目的:观察中药新复方参乌胶囊及其有效成分何首乌二苯乙烯苷对老年大鼠脊髓腰节段神经营养因子表达的影响。方法:雄性SD大鼠,分为青年组(6月龄)、老年组(24月龄)、老年+参乌胶囊小剂量(0.8 g.kg-1)、参乌胶囊大剂量(1.6 g.kg-1)、二苯乙烯苷小剂量(0.03 g.kg-1)、二苯乙烯苷大剂量(0.06 g.kg-1)6组。应用Western blot方法检测大鼠脊髓腰节段神经生长因子(NGF)、脑源性神经营养因子(BDNF)、胶质细胞源性神经营养因子(GDNF)表达。结果:与青年组比较,老年组大鼠脊髓腰节段NGF,BDNF,GDNF的表达量明显减低(P<0.05,P<0.01);灌胃给药3个月,参乌胶囊可显著增加老年大鼠脊髓腰节段的神经营养因子NGF,BDNF,GDNF表达(P<0.05,P<0.01),二苯乙烯苷可明显增加老年大鼠脊髓腰节段GDNF表达(P<0.05)。结论:参乌胶囊及二苯乙烯苷具有提高老年大鼠脊髓腰节段表达神经营养因子的作用,提示该药可能对延缓脊髓神经细胞老化具有潜在的应用价值。
OBJECTIVE: To observe the effect of the new compound Shenwu Capsule and its effective component, stilbene glycosides, on the expression of neurotrophic factors in the lumbar spinal cord of aged rats. METHODS: Male Sprague-Dawley rats were divided into young group (6 months old), old age group (24 months old), old age + Shenwu capsule small dose (0.8 g.kg-1), Shenwu capsule high dose (1.6 g. Kg-1), small dose of stilbene glycoside (0.03 g.kg-1), and large dose of stilbene glycoside (0.06 g.kg-1) 6 groups. Western blot was used to detect the expression of nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF) and glial cell line-derived neurotrophic factor (GDNF) in the lumbar spinal cord of rats. Results: Compared with the young group, the expression levels of NGF, BDNF, and GDNF in the lumbar spinal cord of the old group were significantly decreased (P<0.05, P<0.01). The gavage administration for 3 months significantly increased the old age Wu capsule. The expression of neurotrophic factors NGF, BDNF and GDNF in the lumbar spinal cord of rats was significantly increased (P<0.05, P<0.01). Stilbene glycoside significantly increased the expression of GDNF in the lumbar spinal cord of aged rats (P<0.05). Conclusion: Shenwu Capsule and stilbene glucoside can increase the expression of neurotrophic factor in the lumbar spinal cord of aged rats, suggesting that this medicine may have potential application value in delaying the aging of spinal cord nerve cells.
