目的探讨人突变载脂蛋白E4(apolipoprotein E4,ApoF4)基因表达对转基因鼠血脂代谢的影响。方法应用显微注射法建立人突变ApoE4基因的转基因鼠,分子杂交鉴定。酶法测定人突变ApoE在F1代小鼠染色体上的整合与在血中的表达。酶法测定小鼠血清三酰甘油(triglyceride,TG)和血清胆固醇(cholesterol,TC)水平。结果人突变ApoE4基因稳定地整和于F1代小鼠染色体上并高效表达于血清中。12月龄转基因鼠血脂代谢[血清TG水平(2.89±0.31)mmol/L,血清TC水平(3.02±0.14)mmol/L]与同龄对照组鼠[血清TG水平(2.05±0.12)mmol/L,血清TC水平(2.11±0.01)mmol/L]比较差异有统计学意义(P<0.05,P<0.01)。结论人突变ApoE4基因的异常表达影响到了小鼠的血脂代谢。
Objective To investigate the effect of human mutant apolipoprotein E4 (ApoF4) gene on lipid metabolism in transgenic mice. Methods Transgenic mice with human mutant ApoE4 gene were established by microinjection and identified by molecular hybridization. Enzymatic Determination of Human Mutation ApoE on F1 Generation Mouse Chromosomal Integration and Its Expression in Blood. Enzymatic determination of triglyceride (TG) and serum cholesterol (TC) levels in mice. Results The human mutant ApoE4 gene was stably integrated in the F1 generation mouse chromosome and highly expressed in the serum. Serum TG level (2.05 ± 0.12) mmol / L, serum TC level (3.02 ± 0.14) mmol / L] in the 12-month old transgenic mice were significantly higher than those in the control group Serum TC levels (2.11 ± 0.01) mmol / L] were significantly different (P <0.05, P <0.01). Conclusion The abnormal expression of human mutant ApoE4 gene affects the lipid metabolism in mice.
