目的:研究大鼠灌胃丹参水提物后水溶性成分的吸收情况及在脑组织中的分布情况,建立RP-HPLC法同时测定大鼠血清和脑组织匀浆中丹参水提物三组分丹参素、原儿茶醛和丹参酚酸B的含量。方法:采用液-液萃取法对血清样品和脑组织匀浆样品进行预处理,高效液相色谱内标法测定。ODS Hypersil(4.6 mm×250 mm,5μm)色谱柱,1%乙酸溶液和甲醇采用梯度洗脱,检测波长281 nm。结果:大鼠灌胃丹参水提物后在血清中检测到丹参素、原儿茶醛和丹参酚酸B,在大鼠的脑组织匀浆中未检测到丹参素、原儿茶醛和丹参酚酸B。结论:丹参素、原儿茶醛和丹参酚酸B可经胃肠道吸收入血,本HPLC法稳定、简便、可靠,可用于三者的血药浓度分析。
OBJECTIVE: To study the absorption and distribution of water-soluble components of aqueous extract of Salvia Miltiorrhiza in rats, and to establish a RP-HPLC method for the simultaneous determination of three components of Salvia miltiorrhiza water extracts in serum and brain tissue of rats Danshensu, protocatechuic aldehyde and salvianolic acid B content. Methods: Serum samples and brain homogenate samples were pretreated by liquid - liquid extraction and determined by high performance liquid chromatography. The column of ODS Hypersil (4.6 mm × 250 mm, 5 μm) was eluted with a gradient of 1% acetic acid and methanol. The detection wavelength was 281 nm. Results: Danshensu, protocatechuic aldehyde and salvianolic acid B were detected in the serum of Salvia miltiorrhiza after instilled in the aqueous extract of Radix Salviae Miltiorrhizae. No Danshensu, protocatechuic aldehyde and salvia miltiorrhiza Phenolic acid B. Conclusion: Danshensu, protocatechuic aldehyde and salvianolic acid B can be absorbed into the blood via the gastrointestinal tract. The HPLC method is stable, simple and reliable, and can be used for the analysis of plasma concentration of the three drugs.
