为探明烟碱合成与积累机制,采用大田试验方法,测定云烟87和红花大金元在四川省6个产区烤后烟叶的烟碱含量,检测其在旺长期、现蕾期及成熟期PMT基因的表达量,并分析PMT基因表达量与烟碱含量之间的相关性。结果表明:四川不同产区不同烤烟品种的烟碱含量差异较大,云烟87和红花大金元在古蔺烟区烟碱含量最高,分别为3.40%和4.37%;云烟87在米易烟区烟碱含量最低,为1.27%,红花大金元在会理烟区烟碱含量最低,为1.36%。经相关性分析,2个品种的烟碱含量均与成熟期PMT基因表达量呈极显著正相关,与现蕾期基因表达量呈显著(红花大金元)或极显著(云烟87)正相关。PMT基因表达量是影响烟碱含量的决定因素之一。
In order to explore the mechanism of nicotine synthesis and accumulation, the nicotine content of Yunyan87 and Honghua Dajinyuan in the cured tobacco leaves of six producing areas in Sichuan Province was determined by field test method, and its nicotine content was determined during the long-term, budding and ripening Phase PMT gene expression, and analysis of PMT gene expression and the correlation between nicotine content. The results showed that nicotine content of flue-cured tobacco varieties differed greatly in different flue-cured tobacco varieties in Sichuan Province. The largest content of nicotine in Huahin Daikin was 1.36%. Correlation analysis showed that there was a significant positive correlation between the nicotine content of the two cultivars and PMT gene expression at maturity, which was significantly correlated with the gene expression at budding stage (Honghua Dajinyuan) or extremely significant (Yunyan87) Related. PMT gene expression is one of the determinants of nicotine content.
