目的研究免疫相关基因HLA-G多态性与新疆哈萨克族食管癌易感性的关系。方法采用病例-对照研究方法,调查新疆哈萨克族食管癌患者132例和非食管癌患者254例,应用聚合酶链反应(PCR)技术和聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术分别对HLA-G基因14bp缺失(rs16375)和HLA-G*0105N(rs41557518)多态性进行检测,并比较2个位点不同基因型与食管癌发病风险的关系。结果 rs16375位点基因型为+14 bp/+14 bp、-14 bp/+14 bp、-14 bp/-14 bp;病例组与对照组的基因型及等位基因频率分布差异有统计学意义(2χ=7.54,P=0.02;2χ=6.80,P=0.01);携带-14bp/-14bp基因型的个体发生食管癌的危险性是+14 bp/+14 bp基因型的2.69倍(OR=2.69,95%CI=1.30~5.55);rs41557518位点基因型为C/-、C/C、-/-;病例组与对照组的基因型及等位基因频率分布差异无统计学意义(2χ=1.19,P=0.28;2χ=1.13,P=0.29);同时携带基因型-14bp/-14bp和C/C的个体发生食管癌的危险性是同时携带基因型+14bp/+14bp和C/C个体的2.82倍(OR=2.82,95%CI=1.32~6.04)。结论 HLA-G基因14bp缺失(rs16375)多态性与哈萨克族食管癌发生有关。
Objective To study the relationship between immune-related gene HLA-G polymorphism and susceptibility to esophageal cancer in Kazakh in Xinjiang. Methods A case-control study was conducted in 132 cases of Kazakh esophageal cancer in Xinjiang and 254 cases of non-esophageal cancer patients. Polymerase chain reaction (PCR) and polymerase chain reaction-restriction fragment length polymorphism (PCR- RFLP) were used to detect the polymorphisms of HLA-G gene (rs16375) and HLA-G * 0105N (rs41557518) respectively. The relationship between different genotypes and the risk of esophageal cancer was also compared. Results The genotypes of rs16375 locus were +14 bp / + 14 bp, -14 bp / + 14 bp, -14 bp / -14 bp. There was significant difference in genotype and allele frequencies between case group and control group (2χ = 7.54, P = 0.02; 2χ = 6.80, P = 0.01). The risk of esophageal cancer in individuals carrying -14bp / -14bp genotype was 2.69 times of that of +14 bp / + 14 bp genotype (OR = 2.69, 95% CI = 1.30-5.55). The genotype of rs41557518 was C / -, C / C, - / -. There was no significant difference in genotype and allele frequencies between cases and controls (2χ = 1.19, P = 0.28; 2χ = 1.13, P = 0.29). The risk of esophageal cancer in individuals with genotypes -14bp / -14bp and C / C was also associated with + 14bp / + 14bp and C / 2.82 times of C (OR = 2.82, 95% CI = 1.32 ~ 6.04). Conclusion The 14 bp deletion (rs16375) polymorphism of HLA-G gene is associated with the occurrence of Kazakh esophageal cancer.
