采用~3H-TdR、~3H-脯氨酸、DNA定量检测及MTT比色等方法,研究猪肺动脉缺氧性内皮细胞条件培养液(HECCM)对猪肺动脉平滑肌细胞(PASMC)增生及胶原蛋白合成的影响,并探讨丹参对此过程的阻抑作用。结果表明:HECCM能引起PASMC的线粒体酶活性升高,核内DNA含量增加及~3H-TdR、~3H-脯氨酸掺入量增多。HECCM组PASMC的~3H-脯氨酸掺入量是常氧性内皮细胞条件培养液(NECCM)组的1.83倍。中药丹参注射液能阻抑HECCM引起的PASMC增生和降低HECCM组和NECCM组PASMC的~3H-脯氨酸掺入量。但不影响NECCM组PASMC核的DNA含量及~3H-TdR掺入量。提示缺氧可能会刺激肺动脉内皮细胞(PAEC)合成、分泌某些能促进PASMC增生及胶原蛋白合成的细胞因子,丹参有抑制此过程的效应。
The proliferation of porcine pulmonary arterial smooth muscle cells (PASMC) and collagen synthesis were studied using porcine pulmonary artery hypoxic endothelial cell conditioned media (HECCM) using ~3H-TdR, ~3H-proline, quantitative DNA detection, and MTT colorimetry. The influence of Salvia miltiorrhiza on the inhibition of this process. The results showed that HECCM could increase the mitochondrial enzyme activity of PASMC, increase the nuclear DNA content and increase the incorporation of ~3H-TdR and ~3H-proline. The ~3H-proline incorporation of PASMC in the HECCM group was 1.83 times that of the normoxic endothelial cell conditioned medium (NECCM) group. Chinese medicine Salvia miltiorrhiza injection can inhibit the proliferation of PASMC induced by HECCM and reduce the 3H-proline incorporation of PASMC in the HECCM and NECCM groups. However, it did not affect the DNA content and ~3H-TdR incorporation of PASMC nucleus in NECCM group. It is suggested that hypoxia may stimulate the synthesis of pulmonary artery endothelial cells (PAEC) and secrete certain cytokines that can promote the proliferation of PASMC and collagen synthesis. Salvia miltiorrhiza has the effect of inhibiting this process.
