麻枳降浊方对便秘型肠易激综合征大鼠肠道间质细胞的影响

目的探讨麻枳降浊方治疗便秘型肠易激综合征(IBS-C)的可能作用机制。方法将60只Wistar大鼠随机分为正常组10只和造模组50只,造模组采用冰水灌胃法建立IBS-C模型,将造模成功的48只大鼠随机分为模型组9只、阳性药组10只、高剂量组10只、中剂量组10只、低剂量组9只,阳性药组给予西沙必利片3.6 mg/(kg·d)灌胃,高、中、低剂量组分别给予麻枳降浊方颗粒剂25.2、12.6、6.3 g/(kg·d)灌胃,模型组给予等量蒸馏水灌胃,正常组正常饲养。14天后,HE染色观察肠组织病理变化,免疫组织化学法检测肠组织Cajal间质细胞(ICC)数量和形态的变化,并采用Image-Pro Plus 6.0图像分析系统分析c-kit基因阳性表达面积和积分光密度(IOD)。结果模型组大鼠肠道ICC细胞突起变短,带状、网状结构不明显,各给药组大鼠肠组织ICC数量、形态、分布及网状连接与正常组相似。模型组大鼠c-kit的阳性表达面积及IOD较正常组显著减少(P<0.05)。高、中剂量组和阳性药组c-kit阳性表达面积及IOD较模型组增多(P<0.05),并且高、中剂量组显著高于阳性药组(P<0.05)。结论麻枳降浊方可能通过增加肠组织ICC细胞数量,改善ICC形态结构,增加肠道传导功能,从而改善IBS-C。 Objective To investigate the possible mechanism of Ma Zhi Jiang Zhuo Fang in treating constipation-induced irritable bowel syndrome (IBS-C). Methods Sixty Wistar rats were randomly divided into normal group (n = 10) and model group (n = 50). The model group was given IBS-C model by ice-water gavage. Forty-eight rats were randomly divided into model group 9, positive drug group 10, high dose group 10, medium dose group 10, low dose group 9, positive drug group given cisapride tablets 3.6 mg / (kg · d) intragastric administration, high, Low-dose group were given Mazhutongzhuo-Fang Granule 25.2,12.6,6.3 g / (kg · d) gavage, the model group given equal volume of distilled water, the normal group of normal breeding. Fourteen days later, the pathological changes of intestinal tissue were observed by HE staining. The number and morphology of Cajal interstitial cells (ICC) in intestinal tissue were detected by immunohistochemistry. The positive expression area of ​​c-kit gene was analyzed by Image-Pro Plus 6.0 image analysis system Integral Optical Density (IOD). Results In the model group, the ICC cells in the intestine of the rats in model group were shortened, and the ribbons and reticular structures were not obvious. The number, shape, distribution and reticular connection of ICC in intestinal mucosa in each group were similar to those in normal group. The positive expression area and IOD of c-kit in model group were significantly lower than those in normal group (P <0.05). The c-kit positive area and IOD of high dose group and middle dose group were higher than those of model group (P <0.05), and the levels of c-kit in high dose group and middle dose group were significantly higher than those in positive group (P <0.05). Conclusion Ma Zhijiang Jiangzhuo decoction may improve IBS-C by increasing the number of ICC cells in intestinal tissue, improving the morphological structure of ICC and increasing the intestinal conduction function.
