目的:探讨毛蕊花糖苷对阿尔茨海默病小鼠的神经保护作用。方法:将10月龄APP/PS1双转基因雄性小鼠随机分为模型组,熟地黄低、中、高剂量组(30、60、120 mg·kg-1),同龄C57BL/6小鼠为对照,每组8只。利用Morris法观察各组小鼠学习记忆能力;TUNEL法检测各组神经细胞凋亡情况;Nissl法检测小鼠脑组织神经元尼氏体表达;Western blot法检测各组β淀粉样蛋白表达情况。结果:毛蕊花糖苷中、高剂量组能够显著改善小鼠学习记忆能力;能够增加神经元数量,减少神经细胞凋亡及β淀粉样蛋白沉积。结论:毛蕊花糖苷能够改善阿尔茨海默病小鼠的学习记忆能力,其可能是通过促进神经元存活、减少凋亡并减少Aβ沉积实现的。
Objective: To investigate the neuroprotective effect of verbascoside on Alzheimer’s disease in mice. Methods: The 10-month-old APP / PS1 double transgenic male mice were randomly divided into model group, low, medium and high dose group (30,60,120 mg · kg-1), C57BL / 6 mice , Each group of 8. Morris method was used to observe the learning and memory abilities of mice in each group. TUNEL method was used to detect the apoptosis of nerve cells in each group. Nissl method was used to detect the Nissl body of neurons in the brain tissue of mice. Western blot was used to detect the amyloid beta protein expression in each group. Results: Verbascoside medium and high dose groups could significantly improve the learning and memory ability of mice, increase the number of neurons, reduce the apoptosis of neurons and the deposition of amyloid β protein. Conclusion: Verbascoside can improve learning and memory of Alzheimer’s disease mice, which may be through promoting neuron survival, reducing apoptosis and reducing Aβ deposition.
