研究中药大黄三七散对急性脑出血(ICH)大鼠的神经功能缺损改善的影响并探讨其分子机制。以肝素胶原酶Ⅳ法制作大鼠脑出血模型,考察了施药前后ICH大鼠神经功能评分、脑系数及脑含水量、脑匀浆中自由基释放量及抗氧化酶活力的变化及病理切片观察。结果表明,大黄三七散可明显改善ICH大鼠神经功能缺失症状,显著降低脑系数并减少脑含水量,施加大黄三七散后ICH大鼠脑组织自由基含量明显降低,而机体SOD及CAT抗氧化酶活力提高。改善效果明显呈剂量依赖效应。病理切片观察表明,与模型组相比,大黄三七治疗3个剂量治疗组ICH大鼠的脑组织病变、脑水肿及炎症细胞浸润情况均明显改观,神经细胞损伤亦显著减轻。大黄三七散的解毒、通络及抗氧化功效是改善急性ICH大鼠神经功能缺损的重要分子机制。
To investigate the effect of Rhubarb Sanchisan on the improvement of neurological deficits in acute intracerebral hemorrhage (ICH) rats and its molecular mechanism. The rat model of intracerebral hemorrhage was made by heparin collagenase Ⅳ method. The neurological function score, cerebral coefficient and brain water content of ICH rats before and after administration were measured. The free radical release and antioxidase activity in brain homogenates were measured. Observed. The results showed that rhubarb Sanchi powder could obviously improve the symptoms of neurological deficit in ICH rats, significantly reduce the cerebral coefficient and decrease the water content in the brain. The content of free radicals in brain tissue of ICH rats decreased significantly after the rhubarb Sanchi powder was given, while SOD and CAT Antioxidant enzyme activity increased. The improvement effect was obviously dose-dependent. Pathological observation showed that compared with the model group, rhubarb and Panax notoginseng could significantly improve the pathological changes, brain edema and inflammatory cell infiltration of ICH rats in three dose treatment groups, and significantly reduce the damage of nerve cells. Rhubarb Sanchi powder detoxification, collaterals and anti-oxidant effect is to improve acute ICH rats neurological deficit important molecular mechanism.
