目的 利用体外培养的兔视网膜M櫣ller细胞 ,观察其在高糖、高胰岛素条件下血管内皮生长因子 (VEGF)表达的变化。 方法 采用免疫细胞化学法、原位杂交法和酶联免疫吸附法(ELISA)进行定性定量测定高糖、高胰岛素条件下体外培养的M櫣ller细胞VEGF的表达?〗峁「咛恰⒏咭鹊核啬艽碳櫣ller细胞VEGF的表达 ,并且是通过转录水平调节的 ,高糖对M櫣ller细胞分泌的VEGF的调节呈时间和剂量依赖性。 结论 高糖和高胰岛素都可以通过刺激M櫣ller细胞编码VEGF基因转录 ,增强VEGF蛋白表达 ,而在糖尿病视网膜病变 (DR)的新生血管形成中发挥重要作用。
Objective To observe the changes of vascular endothelial growth factor (VEGF) expression in rabbit retinal Müller cells cultured in vitro under high glucose and high insulin conditions. Methods Immunocytochemistry, in situ hybridization and enzyme-linked immunosorbent assay (ELISA) were used to qualitatively and quantitatively determine the expression of VEGF in M 櫣 ller cells cultured in vitro under high glucose and high insulin condition. 峁 "咛 ⒏ ⒏ 咭The expression of VEGF was significantly up-regulated by transcript levels in high and low glucose groups, and the high glucose had a time-and dose-dependent effect on VEGF secretion in Müller cells.Conclusion Both high glucose and high insulin can be stimulated by stimulation Müller cells encode VEGF gene transcription, enhance VEGF protein expression, and play an important role in neovascularization of diabetic retinopathy (DR).
