目的观察骨桥蛋白(OPN)与整合素αvβ3在主动脉夹层(AD)中的表达及意义。方法雌性C57BL/6小鼠60只随机均分为两组:A组每天腹腔注射血管紧张素Ⅱ4.5mg/kg建立AD模型;B组注射生理盐水作为对照。连续注射14d后处死小鼠,观察AD形成情况,取主动脉组织行HE染色,采用免疫组织化学染色法检测OPN与整合素αvβ3的表达。结果 B组小鼠未形成AD。A组中,6只未见明显AD形成,20只小鼠形成AD,4只小鼠因AD破裂出血死亡。A组OPN和整合素αvβ3阳性表达率均高于B组(86.7%vs.6.7%和80.0%vs.13.3%)(P<0.05)。形成AD小鼠的OPN和整合素αvβ3阳性表达率均高于未形成AD小鼠(95.8%vs.13.9%和87.4%vs.19.4%)(P<0.05)。在形成AD小鼠中,OPN和整合素αvβ3表达呈正相关(rs=0.966,P<0.05)。结论形成AD的小鼠主动脉组织中,OPN与整合素αvβ3呈高表达。
Objective To observe the expression and significance of osteopontin (OPN) and integrin αvβ3 in aortic dissection (AD). Methods Sixty female C57BL / 6 mice were randomly divided into two groups: group A was given intraperitoneal injection of angiotensin II (4.5 mg / kg) daily to establish AD model; group B was injected with saline as control. After 14 days of continuous injection, mice were sacrificed and the formation of AD was observed. The aorta was harvested for HE staining. Immunohistochemical staining was used to detect the expression of OPN and integrin αvβ3. Results Group B mice did not develop AD. In group A, no significant AD was found in 6 of 6 AD mice, and 4 mice died of AD rupture. The positive rates of OPN and integrin αvβ3 in group A were higher than those in group B (86.7% vs.6.7% and 80.0% vs.13.3%, P <0.05). The positive rates of OPN and integrin αvβ3 in AD mice were higher than those in non-AD mice (95.8% vs.13.9% and 87.4% vs.19.4%, respectively) (P <0.05). There was a positive correlation between OPN and integrin αvβ3 expression in AD mice (rs = 0.966, P <0.05). Conclusion The expression of OPN and integrin αvβ3 is highly expressed in the aorta of AD mice.
