目的研究非小细胞肺癌组织中抗凋亡蛋白BAG-1的表达和临床意义,并探讨其与癌细胞凋亡及Bcl-2蛋白表达的关系。方法采用免疫组织化学SP法和TNUEL法分别检测54例非小细胞肺癌组织中BAG-1、Bcl-2蛋白的表达及其细胞凋亡指数,并检测20例正常支气管黏膜组织中BAG-1蛋白的表达作对照。结果非小细胞肺癌BAG-1蛋白表达阳性率为74.07%,明显高于正常支气管黏膜(阳性率为5%)。其表达与非小细胞肺癌组织分化程度、有无淋巴结转移、TNM分期密切相关,而与性别、年龄、病理类型无关。非小细胞肺癌组织中BAG-1蛋白表达与Bcl-2蛋白表达呈正相关关系(Pearson列联系数为0.406,P<0.01)。随BAG-1蛋白表达强度的增强,癌细胞凋亡指数逐渐减少,各组间有统计学意义。结论BAG-1蛋白在非小细胞肺癌组织中表达显著高于正常肺组织,其表达与非小细胞肺癌发生、发展密切相关,与Bcl-2蛋白表达呈正相关,能抑制癌细胞凋亡,可作为肺癌治疗的新靶点。
Objective To study the expression and clinical significance of anti-apoptotic protein BAG-1 in non-small cell lung cancer (NSCLC) and to investigate its relationship with apoptosis and Bcl-2 protein expression. Methods The expressions of BAG-1 and Bcl-2 in 54 NSCLC tissues were detected by immunohistochemical SP method and TNUEL method. The apoptosis index and BAG-1 protein expression were detected in 20 cases of normal bronchial mucosa As control. Results The positive rate of BAG-1 protein in non-small cell lung cancer was 74.07%, which was significantly higher than that in normal bronchial mucosa (the positive rate was 5%). The expression was closely related to the degree of differentiation, lymph node metastasis and TNM stage in non-small cell lung cancer, but not with gender, age and pathological type. There was a positive correlation between BAG-1 protein expression and Bcl-2 protein expression in non-small cell lung cancer (Pearson’s co-ordinate coefficient was 0.406, P <0.01). With the increase of BAG-1 protein expression, the apoptosis index of cancer cells gradually decreased, and there was statistical significance among the groups. Conclusions The expression of BAG-1 protein in non-small cell lung cancer tissues is significantly higher than that in normal lung tissues. The expression of BAG-1 protein is closely related to the occurrence and development of non-small cell lung cancer and positively correlated with the expression of Bcl-2 protein, which can inhibit the apoptosis of cancer cells. As a new target for the treatment of lung cancer.
