目的:探讨清营解表合剂对流感病毒A/PR/8/H1N1感染小鼠免疫保护的作用机制。方法:以亚洲甲型鼠肺适应株A/PR/8/H1N1流感病毒小鼠滴鼻,建立感染模型。分别给予病毒唑、清营解表合剂,计算死亡保护率、延长生命率;测定小鼠胸腺指数和脾脏指数;流式细胞术检测小鼠外周血特异性细胞因子Th1(γ-干扰素,IFN-γ)/Th2(白介素-10,IL-10)表达水平。结果:清营解表合剂组小鼠死亡保护率和生命延长率分别为52.94%和37.64%,小鼠脾脏指数和胸腺指数明显提高。感染后小鼠外周血细胞的Th1型反应弱势表达,Th2细胞因子优势表达,Th1(IFN-γ)/Th2(IL-10)比值明显降低,Th1/Th2表达向Th2偏移。清营解表合剂组Th1细胞因子水平显著升高,且向空白组正常水平方向漂移。结论:清营解表合剂对感染小鼠具有明显的死亡保护作用,能够逆转流感病毒感染小鼠外周血因子Th1向Th2漂移的作用,通过免疫调节作用防治流行性感冒。
Objective: To explore the mechanism of Qingying Jie table mixture on the immunoprotection of influenza A / PR / 8 / H1N1 infected mice. Methods: Infected model was established by intranasal instillation of influenza virus A / PR / 8 / H1N1 in Asian mice. Were given ribavirin, Qingying Jie table mixture, calculate the rate of death protection, to extend the life rate; determination of mouse thymus index and spleen index; flow cytometry detection of mouse peripheral blood specific cytokines Th1 (IFN-γ, IFN -γ) / Th2 (interleukin-10, IL-10) expression levels. Results: The mortality protection rate and life extension rate of mice in Qingying Jiefang Mixture group were 52.94% and 37.64%, respectively. The spleen index and thymus index in mice were significantly increased. Th1 responses were weakly expressed in mouse peripheral blood cells after infection, and the Th2 cytokines were predominantly expressed. The Th1 / IL-10 ratio was significantly decreased and the Th1 / Th2 expression was shifted to Th2. Qingying Jie table mixture of Th1 cytokines levels were significantly increased, and to the normal level of the blank group drifting. CONCLUSION: Qingying Jiexiao Mixture has significant protective effect on infected mice and reverses the effect of Th1 to Th2 shift on the peripheral blood of mice infected with influenza virus. It can prevent and cure influenza by immunomodulation.
