为建立超氧阴离子自由基 (O2·- )引发人红细胞膜氧化损伤实验模型 ,研究马齿苋总黄酮对红细胞膜氧化损伤的保护作用。以邻苯三酚在碱性条件下自氧化产生 (O2·- )引发人红细胞膜氧化损伤 ,并以此为实验模型研究马齿苋总黄酮对红细胞膜封闭能力唾液酸、丙二醛 (MDA)含量的影响。结果显示 :(O2·- )能引起红细胞膜重封闭能力显著降低 ,其平均封闭度为 4 1 9% ,红细胞膜唾液酸含量较正常膜显著下降 ,MDA含量显著增加。而预先加入不同浓度的马齿苋总黄酮后能显著增加膜封闭能力 ,红细胞膜唾液酸含量较氧化膜显著增加 ,MDA含量显著降低。且存在明显的剂量效应依赖关系。提示 :马齿苋总黄酮对红细胞膜氧化损伤具有保护作用。
In order to establish an experimental model of oxidative damage induced by superoxide anion radical (O2·-) in human erythrocyte membrane, the protective effect of total flavonoids from Portulaca oleracea on oxidative damage of erythrocyte membrane was studied. Oxidative damage of human erythrocyte membrane was induced by pyrolysis of pyrogallol under alkaline conditions (O2·-), and as an experimental model, the flavonoids of purslane on erythrocyte membrane sealing ability of sialic acid and malondialdehyde (MDA) were studied. ) The effect of content. The results showed that: (O2·-) can significantly reduce red blood cell membrane re-close ability, the average degree of closure was 41.9%, erythrocyte membrane sialic acid content was significantly lower than the normal membrane, MDA content increased significantly. The addition of different concentrations of total flavonoids of Portulaca oleracea could significantly increase the membrane sealing ability. The content of sialic acid in the erythrocyte membrane was significantly higher than that in the oxide membrane, and the MDA content was significantly reduced. There is a clear dose-effect dependence. Tip: The total flavonoids of Portulaca oleracea have protective effects on oxidative damage of erythrocyte membrane.
