目的:探讨原癌基因cm yb 反义寡核苷酸对脑胶质瘤细胞凋亡的作用及机理。方法:建立C6 脑胶质瘤细胞动物模型,并随机分为三组(每组12 只),用正义、反义cm ybSON和生理盐水瘤区原位注射。动态观察肿瘤的病理组织学改变,并采用流式细胞仪检测各组肿瘤细胞在不同时间及不同对象的凋亡情况。结果:反义SON治疗后的胶质瘤内可见较多炎性细胞浸润和钙化灶;反义治疗组凋亡细胞分别为第4 天34% ,第8 天271% ,第12 天461% ,第16 天484% ,是正义治疗组和对照组相应时段凋亡细胞的35 倍以上(P< 0.05)。结论:cm yb 原癌基因对脑胶质瘤的发生、发展有重要作用;cm yb 反义寡核酸能诱导胶质瘤细胞凋亡,对瘤细胞生长有较明显的抑制作用
Objective: To investigate the effect and mechanism of proto-oncogene cm yb antisense oligonucleotides on apoptosis of glioma cells. METHODS: An animal model of C6 glioma cells was established and randomly divided into three groups (12 in each group) and injected in situ with sense, antisense c-mybSON and saline tumor areas. The histopathological changes of the tumor were dynamically observed, and the apoptosis of the tumor cells at different times and in different subjects was detected by flow cytometry. RESULTS: More inflammatory cell infiltration and calcification were observed in antisense SON-treated gliomas; apoptosis cells in the antisense group were 3.4% on the fourth day and 27.1% on the eighth day, respectively. At days 46.1% and 48.4% at day 16, it was more than 3 times more than the apoptotic cells in the right treatment group and control group (P < 0.05). Conclusion: cm yb proto-oncogene plays an important role in the occurrence and development of gliomas; cm yb antisense oligodeoxynucleotides can induce apoptosis of glioma cells and have a significant inhibitory effect on tumor cell growth.
