目的:观察益气除痞汤对酸性反流性食管炎(RE)模型大鼠食管组织与血浆肿瘤坏死因子-α(TNF-α)及白细胞介素-6(IL-6)水平的影响,探讨益气除痞汤治疗RE的作用机制。方法:60只大鼠随机分成假手术组、模型组、益气除痞汤高剂量组、益气除痞汤低剂量组、多潘立酮组,每组10只。除假手术组外,其他各组大鼠采用不全幽门结扎及食管下括约肌切开术制备酸性反流性食管炎模型,造模第2天,益气除痞汤高、低剂量组、多潘立酮组分别按50 g/kg、10 g/kg、4.5 mg/kg给予相应的药物,假手术组及模型组给予等体积生理盐水,每天分2次灌胃,给药7天。取血液以及食管标本,采用ELISA法测定大鼠食管与血浆中TNF-α以及IL-6的含量。结果:与假手术组比较,模型组大鼠食管组织、血浆中TNF-α及IL-6的含量显著升高,差异均有统计学意义(P<0.05,P<0.01)。与模型组比较,益气除痞汤高、低剂量组、多潘立酮组大鼠食管组织、血浆中TNF-α及IL-6的含量显著降低,差异均有统计学意义(P<0.01)。与益气除痞汤低剂量比较,益气除痞汤高剂量组大鼠食管组织、血浆中IL-6的含量降低的更明显,差异均有统计学意义(P<0.05,P<0.01)。结论:益气除痞汤能够明显降低酸性反流性食管炎模型大鼠食管组织、血浆中TNF-α及IL-6的含量,通过降低TNF-α及IL-6的表达,改善食管局部及外周炎症反应。
Objective: To observe the effects of Yiqi Zaopu Decoction on tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in esophageal tissue and plasma in rats with acidic reflux esophagitis (RE) To explore the mechanism of Yiqi Zhaopang Decoction in treating RE. Methods: Sixty rats were randomly divided into sham-operated group, model group, high-dose Yiqi Zhipu decoction group, low-dose Yiqi Zhipu decoction group and domperidone group, with 10 rats in each group. In addition to the sham operation group, the other groups of rats were treated with incomplete pyloric ligation and esophageal sphincterotomy for the preparation of acid reflux esophagitis model. On the second day after modeling, the rats in high, low, The rats were sacrificed at the dose of 50 g / kg, 10 g / kg and 4.5 mg / kg, respectively. The rats in sham operation group and model group were given equal volume of normal saline. Blood and esophageal specimens were taken for determination of TNF-α and IL-6 in rats’ esophagus and plasma by ELISA. Results: The content of TNF-α and IL-6 in esophageal tissue and plasma in model group was significantly higher than that in sham operation group (P <0.05, P <0.01). Compared with the model group, the content of TNF-α and IL-6 in the esophageal tissue and plasma of rats in the Yiqigatangtang high, low dose and domperidone groups decreased significantly, with statistical significance (P <0.01). Compared with the low dose of Yiqixipi decoction, the content of IL-6 in the esophageal tissue and the plasma of the high-dose Yiqi Zhipi decoction group was significantly lower (P <0.05, P <0.01) . Conclusion: Yiqi Zhaopi Decoction can significantly reduce the levels of TNF-α and IL-6 in the esophageal tissue and plasma of the rats with acid reflux esophagitis and improve the esophagitis by decreasing the expression of TNF-α and IL-6 Peripheral inflammatory reaction.
