目的通过观察牙周炎大鼠牙周组织中骨钙素(bone gla protein,BGP)、白细胞介素6(cytokine interleukin-6,IL-6)水平的变化,来探讨大黄素治疗牙周炎的机制。方法选用100只体重160~200 g Wistar大白鼠,随机分成10组,每组10只。A组:正常对照组。B~J组建立大鼠牙周炎模型。B组:牙周炎对照组。B组在牙周炎造模成功后即与A组一同处死。C、D、E、F组分别于牙周炎模型建立后第1、2、3、4周处死。G、H、I、J组均为大黄素治疗组,分别于治疗后第1、2、3、4周处死,与C~F组形成平行对照。大鼠处死后切取上颌牙周组织块制作标本,评价牙周组织的变化,免疫组织化学方法检测牙周组织中BGP、IL-6的表达。结果大黄素治疗组大鼠牙周组织中BGP的含量在实验的各阶段均高于对照组(P<0.05),IL-6的含量在大黄素治疗第3周后开始低于对照组(P<0.05)。结论大黄素具有促进牙周组织形成、抑制炎性反应的作用,对牙周炎的治疗具有重要意义。
Objective To investigate the changes of bone gla protein (BGP) and interleukin-6 (IL-6) levels in periodontal tissue of periodontitis rats to investigate the effects of emodin on periodontitis. mechanism. Methods One hundred Wistar rats weighing 160-200 g were randomly divided into 10 groups of 10 rats each. Group A: normal control group. Rats with periodontitis were established in groups B~J. Group B: periodontitis control group. Group B was killed together with group A after successful modeling of periodontitis. The C, D, E, and F groups were sacrificed at 1, 2, 3, and 4 weeks after the periodontal model was established. G, H, I, and J were all emodin-treated groups. They were sacrificed at 1, 2, 3, and 4 weeks after treatment, and parallel to the C-F group. After the rats were sacrificed, the maxillary periodontal tissue blocks were cut to make specimens, and the periodontal tissue changes were evaluated. The expression of BGP and IL-6 in the periodontal tissues was detected by immunohistochemistry. Results The content of BGP in the periodontal tissues of emodin-treated rats was higher than that of the control group at each stage of the experiment (P<0.05). The level of IL-6 was lower than that of the control group after the third week of emodin treatment. <0.05). Conclusion Emodin can promote the formation of periodontal tissue and inhibit the inflammatory response, which is of great significance for the treatment of periodontitis.
