目的:探讨HDGF基因特异性siRNA对肝癌细胞系HepG2的增殖能力及ADAM9基因表达的影响。方法:设计并合成HDGF基因特异性的siRNA干扰序列,瞬时转染HepG2细胞。实时荧光定量PCR检测HDGF和ADAM9 mRNA的表达变化;MTT法观察细胞增殖能力的变化。结果:转染48h时HDGF和ADAM9的基因表达抑制最为明显,分别为阴性对照组表达量的7%和5%(P<0.05)。转染48h时HepG2细胞增殖能力受到明显抑制(P<0.05)。结论:HDGF siRNA能够显著抑制HDGF和ADAM9基因的表达,降低HepG2细胞的增殖能力。
Objective: To investigate the effect of HDGF gene-specific siRNA on the proliferation of HepG2 cells and ADAM9 gene expression. METHODS: HDGF gene-specific siRNA interference sequences were designed and synthesized and transiently transfected into HepG2 cells. Real-time fluorescence quantitative PCR was used to detect the expression of HDGF and ADAM9 mRNA. The proliferation of cells was observed by MTT assay. Results: The gene expression inhibition of HDGF and ADAM9 were the most obvious at 48h after transfection, which were respectively 7% and 5% of the negative control group (P <0.05). After 48h, the proliferation of HepG2 cells was significantly inhibited (P <0.05). Conclusion: HDGF siRNA can significantly inhibit the expression of HDGF and ADAM9 genes and reduce the proliferation of HepG2 cells.
