为了进一步探索E类前列腺素(PGE)在免疫调变巨噬细胞系统中可能的作用机理,我们建立了PGE微量放射免疫检测法,应用高比度的~3H-PGE_2以及高度纯化的抗PGE_2抗体,直接定量检测待测样品的PGE含量。此法快速、灵敏、有效、简便。其可测定度在10p8左右,完成整个检测只需4小时左右。此法也可应用于其它种类前列腺素的测定,只须配备相应的抗体以及标准品。体外培养分化成熟的人体激活巨噬细胞仅分泌相当少的PGE,而免疫调变巨噬细胞的PGE分泌量为激活巨噬细胞的50~100倍,提示抑制性巨噬细胞的免疫抑制效应似乎不是PGE介导的。
To further explore the possible mechanism of action of class E prostaglandins (PGEs) in immunomodulatory macrophage system, we established a PGE micro-radioimmunoassay using high specific-3H-PGE_2 and highly purified anti-PGE_2 antibody , Direct quantitative detection of test samples PGE content.
