观察血小板源性生长因子 (PDGF)对于培养人脐静脉内皮细胞 (HUVEC)增殖的影响 ,为血管再生性治疗的靶基因选择提供线索。用流式细胞仪分析细胞周期各时相分布 ,氚 -亮氨酸 ( 3H -Leu)掺入实验评价蛋白质合成 ,免疫组织化学方法检测增殖细胞核抗原 (PCNA)水平。结果不同浓度PDGF均可加速内皮细胞由G1期→S期的转换 ,使细胞进入G2 /M期 ,增加培养细胞3H -Leu掺入量及PCNA水平 ;但PDGF浓度为 5ng时与对照组相比 ,作用不显著。PDGF能够促进二维培养内皮细胞增生 ,具有促血管再生作用
To observe the effect of platelet derived growth factor (PDGF) on the proliferation of cultured human umbilical vein endothelial cells (HUVECs) and to provide clues for the selection of target genes for revascularization. Cell cycle distribution was analyzed by flow cytometry. 3H-Leu incorporation assay was used to evaluate protein synthesis, and proliferating cell nuclear antigen (PCNA) was detected by immunohistochemistry. Results Different concentrations of PDGF could accelerate the transition from G1 phase to S phase of endothelial cells and lead to G2 / M phase and increase 3H-Leu incorporation and PCNA level in cultured cells. When PDGF concentration was 5ng, , The role is not significant. PDGF can promote two-dimensional culture of endothelial cell proliferation, with pro-angiogenic effect
