目的探讨不同白藜芦醇(resveratrol,Res)剂量对大鼠睾丸扭转损伤复位后的超氧化物歧化酶(SOD),过氧化物酶(CAT),丙二醛(MDA),白介素-6(IL-6)及睾丸脑红蛋白(neuroglobin,NGB)的影响。方法 60只成年雄性SD大鼠随机分为扭转复位组为A组;白藜芦醇低剂量(25mg/kg)为B组;白藜芦醇高剂量(50mg/kg)为C组;对照组为(D)组。按Turner法建立单侧睾丸扭转模型。每组根据设定处理方式的不同分别进行处理。扭转复位术后48h,采集左侧睾丸并进行指标(SOD、cAT、MDA及IL-6含量)检测;采用原位缺口末端标记(TUNEL)检测细胞凋亡指数(AI);HE染色及Johnsen评分评估睾丸组织病理改变;电镜观察睾丸组织内生精小管及生精细胞超微改变;RT-PCR法检测睾丸组织中NGB mRNA的表达情况。结果睾丸扭转损伤后,MDA为(2.890±6.5I)mg/L,IL-6为(31.221±7.02)u g/L明显升高,SOD为(240.985±8.19)mg/L,CAT为(1.006±0_31)mg/L及NGB mRNA为(0.35±0.01)明显减少,睾丸组织细胞人量凋亡。应用白藜芦醇后能改善上述改变,且随剂量增大,炎症改变叫显减轻:脑红蛋白基因表达量明显增高,其中A组;B组;C组;D组NGB mRNA含量分别为0.35±0.01;0.42±0.13;0.81±0.19;0.55±0.41,且组间两两比较均有统计学意义(P<0.05)。结论白藜芦醇埘大鼠睾丸扭转具有保护作用,随剂量增中,其保护作用更加显著,同时具有提高抗氧化酶系统的抗氧化及降低血清炎症因子能力,其机制可能是通过诱导NGB mRNA表达而实现。
Objective To investigate the effects of resveratrol (Res) on the levels of superoxide dismutase (SOD), peroxidase (CAT), malondialdehyde (MDA), interleukin-6 IL-6) and testicular neuroglobin (NGB). Methods Sixty adult male Sprague-Dawley rats were randomly divided into group A (group A), group B (25 mg / kg resveratrol), group C (50 mg / kg resveratrol), group C For (D) group. Turner method to establish unilateral testicular torsion model. Each group according to set the different treatment methods. Twenty-eight hours after the reduction, the left testis was collected and the indexes (SOD, cAT, MDA and IL-6 levels) were detected. Apoptotic index (AI) was detected by TUNEL. HE staining and Johnsen score The histopathological changes of testis were evaluated. The ultrastructure of endogenous seminiferous tubules and spermatogenic cells in testis were observed by electron microscope. The expression of NGB mRNA in testis was detected by RT-PCR. Results After testicular torsion injury, MDA was (2.890 ± 6.5) mg / L and IL-6 was significantly higher (31.221 ± 7.02) ug / L and SOD was (240.985 ± 8.19) mg / 0_31) mg / L and NGB mRNA were significantly decreased (0.35 ± 0.01), testicular cells apoptosis. The application of resveratrol can improve the above changes, and with the increase of dose, the change of inflammation was significantly reduced: the expression of neuroglobin increased significantly, of which A group; B group; C group; D group NGB mRNA content was 0.35 ± 0.01; 0.42 ± 0.13; 0.81 ± 0.19; 0.55 ± 0.41, respectively, and there was a significant difference between two groups (P <0.05). CONCLUSIONS: Residual torsion of resveratrol in rats has a protective effect. With the increase of dose, its protective effect is more significant. At the same time, it has the ability to increase antioxidation and reduce serum inflammatory cytokines. The mechanism may be through inducing NGB mRNA Express and achieve.
