【目的】建立广藿香中总黄酮含量的测定方法。【方法】各样品经甲醇提取后,在25℃温度下经聚酰胺柱层析纯化后,分别采用芹菜素—紫外分光光度法和芦丁—亚硝酸钠加硝酸铝络合法于可见光下测定总黄酮含量。【结果】以芹菜素为参照物测得含量的结果为:中国湛江0.83 mg/g、致信药业0.77 mg/g、广东广州0.85 mg/g、越南河内0.94 mg/g、越南河池0.97 mg/g;以芦丁为参照物测得结果为:中国湛江0.81 mg/g,致信药业0.74 mg/g、广东广州0.69 mg/g、越南河内0.73 mg/g、越南河池0.71 mg/g。以芹菜素为参照物测得量高于以芦丁为参照物测得量。【结论】可见分光光度法用于广藿香中总黄酮含量的测定不稳定,紫外分光光度直接测定法则稳定,且快速、简便。
【Objective】 To establish a method for the determination of total flavonoids in patchouli. 【Method】 Each sample was extracted with methanol and purified by polyamide column chromatography at 25 ℃. The samples were determined by visible light spectrophotometry with apigenin-UV spectrophotometry and rutin-sodium nitrite plus aluminum nitrate complexation method respectively Total flavonoids content. 【Result】 The results showed that the content of apigenin was 0.83 mg / g in Zhanjiang, 0.77 mg / g in Zhanjiang, 0.85 mg / g in Guangzhou, Guangdong, 0.94 mg / g in Hanoi, Vietnam, 0.97 mg in Hechi, Vietnam / g; with rutin as reference, the results were as follows: 0.81 mg / g in Zhanjiang of China, 0.74 mg / g in Zhixin Pharmaceutical, 0.69 mg / g in Guangzhou, Guangdong, 0.73 mg / g in Hanoi, Vietnam, 0.71 mg / g in Hechi, . Measured with apigenin as the reference substance was higher than that with rutin as the reference substance. 【Conclusion】 It can be seen that the spectrophotometric method for the determination of total flavonoids in patchouli is unstable and the direct determination of UV spectrophotometry is stable and fast and simple.
