水稻矮缩病毒(rice dwarf virus, RDV)是双壳层的二十面体球形病毒.用含水冷 冻电子显微镜技术和计算机三维重构方法测定去除了外壳层和内部RNA的RDV内壳 层颗粒的3.3nm分辨率的三维结构.结果表明,内壳层表面较为平滑,结构薄而致密, 是RDV的骨架结构.作为内壳层主要结构蛋白的120个P3蛋白质亚基组成60个二聚 体,按T=1的二十面体对称性构成内壳层.内壳层P3亚基的排列与T=13l的外壳层 P8三聚体的排列对照比较表明,P8三聚体与P3亚基是通过非等同并具特征性的方式交 错结合的.
Rice dwarf virus (RDV) is a bilayer icosahedron. The three-dimensional structure of the 3.3 nm resolution of the inner shell particles of the RDV from which the outer shell and the inner RNA were removed was determined using aqueous cryo-electron microscopy and computerized three-dimensional reconstruction. The results show that the inner shell surface is smooth, thin and dense structure, is the RDV skeleton structure. Sixty dimers of 120 P3 protein subunits, which are the major structural proteins of the inner shell, constitute the inner shell with an icosahedral symmetry of T = 1. Comparison of the arrangement of the P3 subunit of the inner shell with the alignment of the trimer of P8 of the outer shell layer with T = 13l shows that the P8 trimer and the P3 subunit are interlaced by a non-identical and characteristic manner.
