目的:分析亳州道地药材紫菀花中槲皮素含量,寻找最佳的提取以及HPLC测定方法。方法:以亳州道地药材紫菀花为对象,考察超声提取以及超高压法提取提取效率,考察溶剂、水解剂、提取时间、提取次数对超声提取法提取率影响,考察溶剂、溶剂浓度、压力、料液比对超高压提取效率影响,选择最佳提取方法,考察流动相、水解温度、盐酸浓度、水解时间对出峰影响,选择最佳的HPLC法,考察不同月份、存储时间对紫菀花中槲皮素含量影响,最后进行定性分析。结果:槲皮素标准品标准曲线Y(峰面积)=8.8985+30.5017X(质量浓度μg/ml),R=0.9998,在0.01~0.5μg/ml范围内维持较高的线性关系;30ml甲醇,25%盐酸10ml,超声提取30分钟,3次提取为最佳提取条件;最佳测定条件高效液相色谱测定:柱温30℃,流动相甲醇:V(0.2%磷酸)=65:35,流速1.0ml/min,波长36nm,参比波长550nm,进样体积10μl;9月份紫菀花中槲皮素含量(7.82±1.31)mg/g高于8月(7.42±1.30)mg/g,新鲜紫菀花槲皮素含量高于1年、1年高于2年,差异具有统计学意义(P<0.05);物质线性范围、检出限以及重现性均可满足需求。结论:HPLC法测定亳州道地药材紫菀花中槲皮素含量效果较好,可作为质量评价工具;亳州道地药材紫菀花中槲皮素达到较高水平,但含量受年份、花期等影响较显著。
Objective: To analyze the content of quercetin in the flower of Asiatic Bozhou, in order to find out the best extraction and HPLC method. Methods: Taking Aster flower of Bozhou medicinal materials as the object, the extraction efficiency by ultrasonic extraction and ultra-high pressure method was investigated. The effects of solvent, hydrolyzate, extraction time and extraction times on extraction rate were studied. The effects of solvent, solvent concentration, The optimum extraction method was selected by the ratio of solid to liquid to the extra-high pressure extraction efficiency. The effects of mobile phase, hydrolysis temperature, hydrochloric acid concentration and hydrolysis time on the peak were investigated. The optimum HPLC method was used to investigate the influence of different storage time Quercetin content, the final qualitative analysis. Results: The standard curve of quercetin standard was Y = 8.8985 + 30.5017X (mass concentration μg / ml), R = 0.9998. The standard curve of quercetin standard maintained a high linearity within the range of 0.01-0.5 μg / ml. 25% hydrochloric acid 10ml, ultrasonic extraction for 30 minutes, three times extraction as the best extraction conditions; the best determination of the conditions of high performance liquid chromatography: column temperature 30 ℃, mobile phase methanol: V (0.2% phosphoric acid) = 65:35, 1.0ml / min, the wavelength of 36nm, the reference wavelength of 550nm, the injection volume of 10μl; September the aster flower quercetin content (7.82 ± 1.31) mg / g higher than August (7.42 ± 1.30) mg / The content of quercetin was higher than 1 year and 1 year was higher than 2 years, the difference was statistically significant (P <0.05). The linear range, detection limit and reproducibility of the material could meet the demand. CONCLUSION: The method of HPLC for the determination of quercetin in the flower of Asiatic Bozhou is better, which can be used as a quality evaluation tool. The quercetin in the flower of Asiatic Bozhou reaches a higher level, but the content is affected by the year, florescence, etc. Significant.
