目的分离原代培养IRM-2小鼠胚胎成纤维细胞(MEFs),从本质上揭示IRM-2小鼠的辐射抗性。方法分离IRM-2小鼠MEFs,并进行原代培养和传代培养。观察MEFs生长形态,测定第3代和第5代细胞生长曲线、细胞周期和增殖指数。结果IRM-2小鼠在胎龄14~16d进行原代MEFs分离,MEFs在体外为贴壁生长型细胞,6代前MEFs生长良好。第3代和第5代MEFs在培养至第3天开始进入对数生长期,到第6天时细胞数达到高峰。MEFs增殖活跃,第3代和第5代细胞增殖指数分别为53%和46%。结论成功分离、培养了IRM-2小鼠MEFs,为进一步利用MEFs研究小鼠的辐射抗性机制奠定了基础。
OBJECTIVE: To isolate primary cultured IRM-2 mouse embryonic fibroblasts (MEFs) and to reveal the inherent radioresistance of IRM-2 mice. Methods MEFs from IRM-2 mice were isolated and cultured in primary culture and subculture. The growth morphology of MEFs was observed and the growth curves, cell cycle and proliferation index of the 3rd and 5th generation cells were determined. Results Primary MEFs were isolated from 14 to 16 days of gestation in IRM-2 mice. MEFs were adherent growth cells in vitro and grew well on the 6th generation. 3rd and 5th generation MEFs began to logarithmic growth phase on day 3 and peaked on day 6. MEFs proliferated actively, and the proliferation indices of 3rd and 5th generation were 53% and 46%, respectively. Conclusion MEFs from IRM-2 mice were successfully isolated and cultured, which laid the foundation for the further study of the mechanism of MEFs on the radiation resistance in mice.
