治疗性抗体在疾病的诊断和治疗方面具有重要意义。鼠源性单克隆抗体的临床应用受限于诱导产生人抗鼠抗体、亲和力低和半衰期短等,随着转基因小鼠和噬菌体展示技术的成熟,人源性治疗性抗体飞速发展,在肿瘤治疗、自身免疫性疾病的治疗、感染性疾病的治疗及减轻移植排斥反应、体内定位诊断、免疫调节等方面发挥重要作用。本文综述了治疗性抗体在肿瘤及免疫、移植等疾病中的治疗应用及人源性治疗性抗体的制备。
Therapeutic antibodies are of great importance in the diagnosis and treatment of diseases. The clinical application of murine monoclonal antibodies is limited to the induction of human anti-mouse antibodies, with low affinity and short half-life, with the development of transgenic mice and phage display technology, the rapid development of human therapeutic antibodies, tumor therapy , The treatment of autoimmune diseases, the treatment of infectious diseases and the reduction of transplant rejection, in vivo diagnosis, immunomodulation and other aspects play an important role. This review summarizes the therapeutic applications of therapeutic antibodies in diseases such as oncology and immunology, transplantation and the preparation of human therapeutic antibodies.
