目的初步探讨不同的胚胎活检方法对囊胚的形成和妊娠结局的影响。方法对14例共17个周期行卵胞浆内单精子注射(ICSI)受精后的D3的4-10细胞胚胎进行活检,吸取1~2个卵裂球细胞做PCR分析,在D5或D6选择合适的胚胎移植。比较新鲜胚胎和解冻胚胎活检后的囊胚形成率及机械法开口与激光法开口的囊胚形成率、临床结局。结果17个活检周期共获卵297个,活检胚胎数207个,164个检出结果,结果检出率79.2%。活检后胚胎的囊胚形成数为106个(51.2%)。移植15个周期,4例获妊娠,临床妊娠率26.6%。新鲜胚胎活检后的囊胚形成率较解冻胚胎的高(分别为61.6%和37.5%),机械法活检后的囊胚形成率和妊娠率较激光法的高(分别是59.66%,44.4%和39.77%,0),两组均有显著性差异。结论机械法活检可获得较激光法更好的囊胚形成率和妊娠结局。新鲜胚胎活检后的囊胚形成率较解冻后的高。
Objective To investigate the effect of different embryo biopsy methods on blastocyst formation and pregnancy outcome. Methods Fourteen of 14 embryos with D3 cycles after intracytoplasmic sperm injection (ICSI) of 17 cycles were biopsied. One or two blastomere cells were harvested for PCR analysis. After D5 or D6 selection Appropriate embryo transfer. The rate of blastocyst formation after biopsy of fresh embryos and thawed embryos was compared, and the blastocyst formation rate and clinical outcome of mechanical opening and laser opening were compared. Results A total of 297 eggs were obtained in 17 biopsies, 207 biopsies and 164 detected results. The detection rate was 79.2%. The number of blastocysts formed after biopsy was 106 (51.2%). 15 cycles of transplantation, 4 cases of pregnancy, the clinical pregnancy rate was 26.6%. The blastocyst formation rate of fresh embryo biopsies was higher than thawed embryos (61.6% and 37.5%, respectively). The blastocyst formation rate and pregnancy rate after mechanical biopsy were higher than those of laser method (59.66%, 44.4% and 39.77%, 0), there are significant differences between the two groups. Conclusion Mechanical biopsy can get better blastocyst formation rate and pregnancy outcome than laser. The blastocyst formation rate after fresh embryo biopsy was higher than that after thawing.
