目的 研究Ⅱ型糖尿病患者脂肪组织胰岛素受体底物 1(IRS 1)及含 2个SH2 结构的蛋白酪氨酸磷酸酶 (SH PTP2 )蛋白表达 ,探讨脂肪组织产生胰岛素抵抗的分子机制。方法 用Western杂交检测Ⅱ型糖尿病患者腹部皮下与大网膜脂肪组织IRS 1、SH PTP2 的蛋白表达 ;同时比较患者自身皮下与大网膜内蛋白表达水平。结果 IRS 1蛋白表达 (A值 )在糖尿病患者皮下脂肪 (2 14± 0 6 7)、大网膜内 (3 2 5± 0 70 )均较对照组 [4 33± 0 5 7(P <0 0 0 1)、8 6 5± 2 85 (P <0 0 5 ) ]减少 ;而SH PTP2与对照组比较差异无显著性 (P >0 0 5 )。IRS 1蛋白表达在患者自身皮下脂肪 (1 0 9± 0 13)较大网膜(2 10± 0 2 2 )减少 (P <0 0 5 ) ;而SH PTP2 在皮下 (70 75± 2 18)则较大网膜 (4 3 6 9± 11 0 7)增加 (P <0 0 5 )。结论 Ⅱ型糖尿病患者脂肪组织IRS 1及SH PTP2 蛋白表达的异常改变可能是引起其产生胰岛素抵抗的机制之一 ;Ⅱ型糖尿病患者的腹部皮下脂肪和内脏脂肪一样存在胰岛素抵抗。
Objective To investigate the expression of insulin receptor substrate 1 (IRS 1) and SHPTP2 protein with two SH2 structures in type 2 diabetic patients and to explore the molecular mechanism of adipose tissue insulin resistance. Methods Western blotting was used to detect the protein expression of IRS 1 and SH PTP2 in subcutaneous and omental adipose tissue of type Ⅱ diabetic patients. Meanwhile, the protein expression of subcutaneous and omental in patients with type Ⅱ diabetes mellitus was compared. Results The expression of IRS 1 protein in subcutaneous fat (2 14 ± 0 6 7) and omentum (3 2 5 ± 0 70) in diabetic patients was significantly higher than that in control group [437 ± 0 57 (P <0) 0 0 1) and 8 6 5 ± 2 85 (P 0 05), respectively. There was no significant difference between SHPTP2 and control group (P 0 05). The expression of IRS-1 protein in patients with subcutaneous fat (109 ± 0 13) was lower than that in the omentum (2 10 ± 0 2 2) (P <0 05), while SH PTP2 was subcutaneous (70 75 ± 2 18) The greater omentum (4396 ± 1170) increased (P <0.05). Conclusions Abnormal changes of IRS 1 and SH PTP2 protein expressions in type 2 diabetic patients may be one of the mechanisms that cause insulin resistance. Type 2 diabetes patients have the same insulin resistance in abdominal subcutaneous fat and visceral fat.
