目的探讨乳腺癌组织乳腺癌易感基因1(BRCA-1)蛋白表达与原癌基因人类表皮生长因子受体2(HER-2)的基因扩增关系。方法分别采用免疫组化法和荧光原位杂交法检测84例乳腺癌(导管内癌32例,浸润性导管癌36例,纤维腺瘤16例)组织中的BRCA-1蛋白表达和HER-2基因扩增情况。结果 BRCA-1蛋白阳性表达63例,阳性表达率为75.0%(63/84)。BRCA-1蛋白表达与淋巴结转移有关(P<0.05)。与不同组织类型无关(P>0.05)。HER-2基因扩增阳性40例,HER-2基因扩增阴性44例。36例有淋巴结转移患者HER-2扩增阳性率为86.1%(31/36),明显高于48例无淋巴结转移患者的27.1%(13/48)(P<0.01)。HER-2基因扩增阳性组BRCA-1蛋白阳性表达率为90.0%(36/40),高于HER-2基因扩增阴性组的22.7%(10/44)(P<0.01)。结论 BRCA-1蛋白表达、HER-2基因扩增与乳腺癌的发生发展有关,BRCA-1蛋白表达与HER-2基因扩增可能存在相关性。
Objective To investigate the relationship between the expression of BRCA-1 in breast cancer and the gene amplification of proto-oncogene human epidermal growth factor receptor 2 (HER-2). Methods Immunohistochemistry and fluorescence in situ hybridization were used to detect the expression of BRCA-1 protein in 84 cases of breast cancer (32 cases of intraductal carcinoma, 36 cases of invasive ductal carcinoma and 16 cases of fibroadenoma) and the expression of HER-2 Gene amplification. Results The positive expression rate of BRCA-1 protein in 63 cases was 75.0% (63/84). The expression of BRCA-1 protein was correlated with lymph node metastasis (P <0.05). Not related to different types of tissues (P> 0.05). HER-2 gene amplification in 40 cases, HER-2 gene amplification in 44 cases. The positive rate of HER-2 amplification in 36 patients with lymph node metastasis was 86.1% (31/36), which was significantly higher than that in 48 patients without lymph node metastasis (27.1%, 13/48) (P <0.01). The positive rate of BRCA-1 protein expression in HER-2 positive group was 90.0% (36/40), which was higher than that in HER-2 negative group (22.7%, 10/44) (P <0.01). Conclusion The expression of BRCA-1 protein and the amplification of HER-2 gene are related to the occurrence and development of breast cancer. The expression of BRCA-1 protein may be correlated with the amplification of HER-2 gene.
