目的观察HIV病毒对不同温度的敏感性,为HIV消毒和灭活提供实验依据。方法制备HIV-1SF33病毒上清,分别放置在4℃冰箱,25℃、37℃、56℃和70℃水浴中,间隔一定时间后取样测定病毒滴度(TCID50)。结果HIV-1SF33滴度在4℃、25℃和37℃下分别于12h、6h和4h后出现10倍左右的降低,且随着时间的延长,HIV-1SF33滴度下降趋势越为明显;随着温度的升高,HIV-1SF33滴度随之下降,4℃和25℃条件下,HIVSF33放置24h,滴度分别下降2.7log和3.7log,37℃条件下放置24h,病毒丧失感染性;56℃作用10min或70℃作用2min,HIV病毒被迅速灭活,已检测不出活病毒。结论HIV病毒对热敏感,但在4℃和25℃条件下,液体环境中的HIV病毒可存活一段时间,应重视冷藏和常温下HIV污染物的消毒。热处理是灭活HIV病毒的有效方法。
Objective To observe the sensitivity of HIV virus to different temperatures and provide experimental evidence for disinfection and inactivation of HIV. Methods The supernatant of HIV-1SF33 virus was prepared and placed in 4 ℃ refrigerator, 25 ℃, 37 ℃, 56 ℃ and 70 ℃ water bath, and the virus titer (TCID50) was measured after sampling for a certain time. Results The titer of HIV-1SF33 tenfoldly decreased at 12h, 6h and 4h at 4 ℃, 25 ℃ and 37 ℃, respectively, and the titer of HIV-1SF33 tended to decrease with the extension of time. The HIV-1SF33 titer decreased with increasing temperature. HIVSF33 was stored at 4 ℃ and 25 ℃ for 24h, and the titer decreased by 2.7log and 3.7log, respectively. After incubation at 37 ℃ for 24 hours, the virus lost its infectivity. 56 ℃ role of 10min or 70 ℃ role 2min, HIV virus is rapidly inactivated, has not been detected live virus. Conclusion The HIV virus is sensitive to heat. However, under the condition of 4 ℃ and 25 ℃, the HIV virus in the liquid environment can survive for a period of time, and the disinfection of HIV contaminants should be emphasized during cold storage and normal temperature. Heat treatment is an effective way to inactivate the HIV virus.
