目的探讨IFRD1表达下调后人肝癌细胞SMMC-7721基因表达谱的改变。方法采用脂质体lipo2000将构建的含目的基因shRNA的质粒转染入SMMC-7721细胞,并应用Affymetrix U133 plus 2.0 array基因芯片检测目的基因表达下调后,该细胞基因表达谱的变化。结果 RNA干扰抑制IFRD1基因表达后,基因芯片检测发现25476个基因中,表达量发生明显改变的有4221个(上下调大于等于1.5倍),上调的基因有1109个,下调的有3 112个;这些基因涉及到细胞周期、细胞凋亡、DNA复制等通路。结论 IFRD1基因表达下调后,人肝癌细胞SMMC-7721基因表达谱发生明显变化,与细胞生长增殖关系密切的细胞信号通路也发生了明显的改变。
Objective To investigate the gene expression profile of human hepatoma SMMC-7721 cells after the down-regulation of IFRD1 expression. Methods The plasmid containing the target gene shRNA was transfected into SMMC-7721 cells with Lipofectamine 2000. The Affymetrix U133 plus 2.0 array gene chip was used to detect the gene expression profile of the target gene after it was down-regulated. Results After RNA interference inhibited the expression of IFRD1 gene, 4221 genes (up - regulated more than or equal to 1.5 times) were found in 25476 genes. The number of up - regulated genes was 1109 and down - regulated was 3 112. These genes involve the cell cycle, apoptosis, DNA replication and other pathways. Conclusion The downregulation of IFRD1 gene expression in human hepatocellular carcinoma cells significantly changes the gene expression profile of SMMC-7721 cell proliferation and cell signaling pathways have also been significantly changed.
