目的:研究不同炮制条件下黄芥子中芥子碱硫氰酸盐含量的变化。方法:采用高效液相色谱法,Kromasil C18(4.6mm×250mm,5μm)色谱柱,流动相为乙腈-0.08mol/L磷酸二氢钾溶液(12∶88),检测波长326nm。结果:芥子碱硫氰酸盐在0.8~4μg范围内具有良好的线性关系,相关系数r=0.9997,平均加样回收率99.23%,RSD为3.12%(n=5)。结论:随着炮制温度的提高和炮制时间的延长,黄芥子中芥子碱硫氰酸盐的含量不断降低,其最佳炮制条件为140℃、5~10min。
Objective: To study the change of thiocyanate content in Sinorhizobium hippophae under different processing conditions. Methods: Kromasil C18 column (4.6 mm × 250 mm, 5 μm) was used with mobile phase of acetonitrile-0.08 mol / L potassium dihydrogen phosphate solution (12:88) at a detection wavelength of 326 nm. The results showed that there was a good linear relationship between the concentration of sinapine and thiocyanate in the range of 0.8 ~ 4μg, the correlation coefficient was 0.9997, the average recovery was 99.23%, RSD was 3.12% (n = 5). CONCLUSION: The content of thiocyanate in Sinorhizobium mustardum decreased with the increase of processing temperature and processing time, and the optimum processing conditions were 140 ℃, 5 ~ 10min.
