目的 :探讨聚合酶链反应 (PCR)扩增大片段线粒体DNA方法。方法 :快速碱裂解法提取线粒体DNA ,长时程PCR扩增出 5 430bp线粒体DNA。结果 :扩增出 5 430bp线粒体DNA产物特异性强 ,产量高。 结论 :该方法反应条件简便、稳定 ,也可用于其它大片段的扩增 ,有一定的实用价值。
Objective: To explore the method of polymerase chain reaction (PCR) amplification of large fragment mitochondrial DNA. Methods: Mitochondrial DNA was extracted by rapid alkaline lysis and 5 430 bp mitochondrial DNA was amplified by long-term PCR. Results: The 5 430bp mitochondrial DNA product was amplified with high specificity and high yield. Conclusion: The method has the advantages of simple and stable reaction conditions, and can also be used for amplification of other large fragments, and has certain practical value.
