目的拟构建气虚痰浊证高血压大鼠模型,为中医药研究提供载体。方法将50只12周龄自发性高血压大鼠(spontaneously hypertensive rats,SHRs),随机分为模型组(40只)及对照组(10只),同时设置10只12周龄正常血压大鼠(wistar-Kyoto,WKY)为对照(WKY组)。模型组隔日喂食高脂饲料,每周6次力竭游泳;对照组及WKY组始终以普通大鼠饲料喂养。对各组大鼠表征体系进行动态观察,12周后进行相关指标检测。结果模型组大鼠随着造模时间延长,分别出现生物学表征评分升高、游泳时间缩短、体质量增加、旷场分析中央格停留时间延长、水平运动距离缩短、血压升高等;血清指标检测中,模型组甘油三酯(TC)、胆固醇(TG)、血糖(GLU)均高于其他2组(P<0.01);模型组低密度脂蛋白(LDL-C)较WKY组升高(P<0.05);模型组双侧颈动脉血流量比对照组、WKY组降低,差异有统计学意义(P<0.01)。结论以SHRs为基础,持续力竭游泳、隔日禁食、高脂饲料喂养建立气虚痰浊证高血压大鼠模型,相关表征及血清指标改变符合气虚痰浊证候特点。
Objective To construct a rat model of Qi-deficiency and phlegm-turbidity hypertension and provide a carrier for the study of traditional Chinese medicine. Methods Fifty 12-week-old spontaneously hypertensive rats (SHRs) were randomly divided into model group (n = 40) and control group (n = 10) wistar-Kyoto, WKY) as a control (WKY group). The model group fed high fat diet on alternate days, and exhausted swimming six times a week. The control group and WKY group were fed with ordinary rat feed. Each group of rat characterization system for dynamic observation, 12 weeks after the relevant indicators. Results The rats in model group showed higher scores of biological characterization, shorter swimming time, increased body weight, longer stay time in open field analysis, shorter horizontal movement distance and higher blood pressure as the modeling time prolonged. Serum indexes (TC), cholesterol (TG) and blood glucose (GLU) in the model group were higher than those in the other two groups (P <0.01). LDL-C in the model group was higher than that in the WKY group <0.05). The bilateral carotid blood flow in model group was lower than that in control group and WKY group (P <0.01). Conclusion SHRs-based, sustained exhaustive swimming, fasted every other day, high-fat diet to establish qi deficiency and phlegm turbid hypertensive rat model, the relevant characteristics and changes in serum indicators in line with the characteristics of qi deficiency and phlegm syndrome.
