以抗寒与寒敏感狗牙根为材料,用12对EST-SSR、SSR引物分别对12份抗寒与寒敏感狗牙根(已通过表型、生理鉴定)进行扩增。结果表明:EST-SSR引物(ES3862、ES8174、ES6865)、SSR引物(CDE45、CDE33、CDE95)可将抗寒及寒敏感狗牙根区分开;EST-SSR引物组合(ES5349+ES3862、ES5349+ES8174、ES2010+ES5349)、SSR引物组合(CDE55+CDE45)能将12份材料区分开,最终构建了可以区分抗寒种质的数字图谱。利用标记获得的0、1数字矩阵对材料进行亲缘关系分析的结果表明:两类标记分别扩增出38条、44条带,多态性比例为97.43%、95.65%;各材料遗传相似系数分别在0.4858~0.9487、0.4855~1.000之间,在遗传相似系数为0.4858和0.4855处能将抗寒与寒敏感狗牙根明显区分。
Cold-resistant and cold sensitive bermudagrass were used as materials. Twelve pairs of EST-SSR and SSR primers were used to amplify 12 cold and cold sensitive bermudagrass (phenotypic and physiological identification) respectively. The results showed that EST-SSR primers (ES3862, ES8174, ES6865) and SSR primers (CDE45, CDE33 and CDE95) could distinguish cold and cold susceptible Bermudagrass. EST- SSR primer combinations (ES5349 + ES3862, ES5349 + ES8174, ES2010 + ES5349), SSR primer combinations (CDE55 + CDE45) can distinguish 12 materials and finally construct a digital map that can distinguish cold-resistant germplasms. The results of genetic relationship analysis using 0,1 digital matrix obtained from the markers showed that 38 bands and 44 bands were amplified respectively by the two types of markers, with the proportions of polymorphisms of 97.43% and 95.65%, respectively. The genetic similarity coefficients of the materials Between 0.4858 ~ 0.9487,0.4855 ~ 1.000, the genetic similarity coefficient of 0.4858 and 0.4855 at cold-sensitive and cold sensitive bermudagrass can be clearly distinguished.
