目的:利用傅里叶变换红外光谱结合聚类分析法对不同加工方式肉苁蓉的宏观差异进行分析,为名贵濒危中药肉苁蓉加工方式的鉴别和选择提供方法和依据。方法:通过采用带有全反射衰减附件(ATR)的红外光谱仪,在4000~650 cm~(-1)波段内检测3种整株加工和5种不同切片加工方式处理的180个肉苁蓉样品粉末,得到红外光谱图,图谱通过ATR校正及预处理后,应用Assure ID软件进行聚类分析。结果:以识别率和拒绝率为指标判断聚类结果,当两个值都越大时,表明这两类物质之间没有重叠,可以较好地将其聚类然后分开。整株加工和切片加工的药材样品分别聚类,结果显示整株加工的肉苁蓉样品中,3种加工方式的识别率和拒绝率最高值均达到100%,最低值为76%;切片加工的肉苁蓉样品中,识别率最高达100%,拒绝率最低为72%。结论:红外光谱结合聚类分析不仅能有效区分不同加工方式的肉苁蓉样品,并且能佐证产地加工对药材成分变化的影响,为加工炮制药材的快速鉴别和质量控制提供了方法。
OBJECTIVE: To analyze the macroscopic differences of Cistanche tuberculosis by different processing methods by using Fourier transform infrared spectroscopy and cluster analysis, and to provide methods and basis for the identification and selection of Cistanche tubulosa (Cistanche tubulosa) processing of rare and endangered Chinese medicines. Methods: Three Cistanche deserticola samples with three whole processing and five different slice processing methods were detected in 4000 ~ 650 cm ~ (-1) band by using infrared spectrometer with total reflection attenuation attachment (ATR) The infrared spectrum was obtained. After the spectrum was corrected by ATR and pretreated, the Assure ID software was used for cluster analysis. Results: Judging the clustering results based on the recognition rate and rejection rate, when both values are larger, it shows that there is no overlap between the two types of materials, which can be better clustered and then separated. The results showed that the highest recognition rate and rejection rate of the three kinds of processing methods all reached 100% and the lowest value was 76%. Cistanche In the sample, the recognition rate is up to 100% and the rejection rate is at least 72%. Conclusion: The combination of infrared spectroscopy and cluster analysis can not only effectively distinguish the cistanche samples from different processing methods, but also prove the influence of the processing of origin on the changes of the components of the medicinal materials, providing a method for the rapid identification and quality control of the processed medicinal herbs.
