目的:探讨合适的饲料配方,在较短的实验周期内形成稳定、适用的高脂高同型半胱氨酸血症(hyperhomocysteinemia,HHcy)动物模型。方法:40只雄性SD大鼠分为4组,每组10只,分别按组给予普通饲料、配方1(基础饲料占88.5%,高脂高胆固醇不添加丙硫氧嘧啶)、配方2(较配方1增加5%蛋黄糖,5%全脂奶粉,0.15%丙硫氧嘧啶)、配方3(0.25%丙硫氧嘧啶,较配方2增加5%蛋黄粉)饲料常规喂养。每周称1次体重,观察体重变化。分别于2,4周取血测定血清中总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)变化及血浆同型半胱氨酸(homocysteine,Hcy)的浓度。结果:与对照组相比配方1组体重没有明显差异;配方2组,3组体重显著下降(P<0.01)。2,4周时与对照组相比,血清TC和LDL-C含量及血浆Hcy浓度,配方1组没有显著性差异;配方2组,3组都有显著升高(P<0.01)。结论:该实验研究调整后的大鼠饲料配方2组是比较合适的饲料配方组成,能在较短的时间内形成稳定的大鼠高脂HHcy血症动物模型。
OBJECTIVE: To explore a suitable feed formulation to form a stable and suitable hyperhomocysteinemia (HHcy) animal model in a short experimental period. Methods: Forty male Sprague-Dawley rats were divided into 4 groups with 10 animals in each group. The animals were fed with normal diet, Formula 1 (88.5% basal diet, high-fat and high cholesterol did not add propylthiouracil), formula 2 Formula 1 added 5% egg yolk sugar, 5% whole milk powder, 0.15% propylthiouracil), formula 3 (0.25% propylthiouracil, formula 2 increased 5% egg yolk powder) feed routine feeding. Weighed weekly weighing 1, observed body weight changes. The levels of total cholesterol (TC), low density lipoprotein cholesterol (LDL-C) and plasma homocysteine (Hcy) in serum were determined at 2 and 4 weeks respectively. Results: Compared with the control group, there was no significant difference in the body weight between the two groups. The body weight of the second and third groups decreased significantly (P <0.01). At 2 and 4 weeks, the levels of TC and LDL-C in serum and plasma Hcy concentrations in the two groups had no significant difference (P <0.01). Conclusion: This experimental study of the adjusted rat feed formula 2 is more appropriate feed composition, in a relatively short period of time to form a stable animal model of hyperlipidemic HHcymia in rats.
