目的:探讨山楂叶总黄酮对大鼠心肌缺血性损伤的保护作用及通过HO-1诱导途径的作用机制。方法:建立皮下注射异丙肾上腺素造成大鼠心肌缺血模型,观察山楂叶总黄酮对心电图、血清肌酸激酶CK)和乳酸脱氢酶(LDH)活性、心肌超氧化物歧化酶(SOD)和还原型谷胱甘肽(GSH)的活性以及血红素氧合酶-1(HO-1)蛋白表达和mRNA水平的影响。结果:山楂叶总黄酮(100~200mg/kg)能显著对抗异丙肾上腺素所致急性大鼠心肌缺血心电图T波的异常,显著降低血清LDH和CK活力,增加心肌组织SOD和GSH活性,上调HO-1的表达,并呈一定的剂量依赖性。HO-1抑制剂组能够部分取消HLF的对异丙肾上腺素造成的心肌损伤的保护作用。结论:山楂叶总黄酮对急性缺血性心肌损伤的保护作用机制与诱导HO-1的表达密切有关。
Objective: To investigate the protective effect of hawthorn leaves flavonoids on myocardial ischemic injury and the mechanism of its induction by HO-1. Methods: A rat model of myocardial ischemia was established by subcutaneous injection of isoproterenol. The effects of hawthorn leaf flavonoids on electrocardiogram, serum creatine kinase (CK) and lactate dehydrogenase (LDH) activity, myocardial SOD activity, And glutathione (GSH) activity as well as heme oxygenase-1 (HO-1) protein expression and mRNA levels. Results: Total flavonoids of hawthorn leaves (100 ~ 200mg / kg) could significantly antagonize T wave abnormalities in acute myocardial ischemia induced by isoproterenol, significantly reduce serum LDH and CK activity, increase myocardial SOD and GSH activity, Up-regulated the expression of HO-1, and showed a dose-dependent manner. The HO-1 inhibitor group partially abolished the protective effect of HLF on isoproterenol-induced myocardial injury. Conclusion: The protective effect of hawthorn leaf flavonoids on acute ischemic myocardial injury is closely related to the induction of HO-1 expression.
