目的对体外炎性环境下小鼠成肌细胞/分化肌管的免疫学特性进行检测,分析其是否具备抗原呈递功能。方法用IFN-γ刺激小鼠成肌细胞(C_2C_(12))及马血清诱导分化后的肌管,通过免疫荧光染色、q-PCR、Western blot、流式细胞术检测细胞表面H-2k~b、MHC-II、TLR3及相关细胞因子水平的改变。结果 IFN-γ刺激后,免疫荧光染色、q-PCR、Western blot均检测到成肌细胞/分化肌管表面MHC分子表达上调,Western blot检测到TLR3表达上调,q-PCR检测到细胞因子IL-1β、IL-6、IL-10、IL-18、MCP-1及TGF-β表达上调。结论在炎性条件下,成肌细胞/分化肌管具备炎性细胞表征,具备抗原呈递功能。
Objective To detect the immunological characteristics of mouse myoblasts / myotubes in inflammatory environment in vitro and analyze whether they possess antigen presenting function. Methods The myotubes induced by IFN-γ were stimulated with C_2C_ (12) and horse serum. The expression of H-2k ~ b, changes in MHC-II, TLR3 and related cytokines. Results After induction of IFN-γ, the expression of MHC on myoblast / myotube surface was up-regulated by immunofluorescence, q-PCR and Western blot. The expression of TLR3 was up-regulated by Western blot. The levels of IL- 1β, IL-6, IL-10, IL-18, MCP-1 and TGF-β. Conclusion Under inflammatory conditions, myoblasts / differentiated myotubes possess inflammatory cell characterization and antigen presenting function.
