在单因素试验的基础上,采用正交试验的方法,对水煎煮法和酶解法提取沙棘叶中绿原酸的工艺条件进行了优化。试验结果表明:水煎煮法最佳提取工艺条件为:料液比(g∶mL)1∶35,煎煮时间1.0 h,煎煮次数3次。该条件下,沙棘叶中绿原酸提取率为2.42%。酶解法最佳提取工艺条件为:以加5%纤维素酶的pH 4.5的酸性水为提取剂,料液比(g∶mL)1∶15,温度50℃,提取时间1.0 h。该条件下,绿原酸提取率为3.49%。酶解法与水煎煮法相比,绿原酸提取率提高了44.2%。
On the basis of single factor experiment, the orthogonal experiment was used to optimize the extraction process of chlorogenic acid in seabuckthorn leaves by decocting method and enzymolysis method. The results showed that the optimal extraction conditions were as follows: the ratio of material to liquid (g: mL) 1:35, boiling time 1.0 h, boiling times 3 times. Under this condition, the extraction rate of chlorogenic acid in seabuckthorn leaves was 2.42%. The optimum conditions of enzymatic hydrolysis were as follows: acidic water (pH 4.5) supplemented with 5% cellulase was used as extraction solvent, the ratio of solid to liquid (g: mL) was 1:15, the temperature was 50 ℃ and the extraction time was 1.0 h. Under these conditions, the extraction rate of chlorogenic acid was 3.49%. Enzymolysis method compared with the decoction method, chlorogenic acid extraction rate increased by 44.2%.
