应用S—P免疫组织化学方法研究62例(男性51例,女性11例,年龄34~68岁,平均年龄53.7岁)肺癌组织中P16蛋白和P53蛋白的表达情况,并进行增殖细胞核抗原(PCNA)检测,计算细胞增殖指数(Proliferationindex,PI)。62例肺癌组织中P16蛋白和P53蛋白阳性率分别为58.1%和59.7%。腺癌P16蛋白的阳性率明显高于小细胞癌(P<0.05);淋巴结转移阳性组P16蛋白的表达显著低于阴性组(P<0.05);PI分级为Ⅱ级的P16蛋白表达显著高于Ⅳ级(P<0.05)。不同组织类型肺癌中P53蛋白的表达未见明显差异,淋巴结转移阳性组P53蛋白的表达高于阴性组(P〈0.01);不同PI分级中P53蛋白的表达,Ⅳ级明显高于Ⅰ级(P〈0.05)和Ⅱ级(P〈0.05),Ⅲ级明显高于Ⅰ级(P〈0.05)和Ⅱ级(P〈0.05)。P16蛋白低表达和P53蛋白过表达之间未见明显相关。揭示P16蛋白低表达和P53蛋白过表达均有促进肺癌细胞增殖的作用,P16蛋白的表达与肺癌的细胞分化有关,P53蛋白过表达对肺癌细胞的转移起重要作用。抑癌基因P53对MTS1/P16基因无明显调控作用,检测P53蛋白表达?
S-P immunohistochemical method was used to study the expression of P16 protein and P53 protein in 62 cases (51 males, 11 females, aged 34-68 years, mean age 53.7 years) and performed proliferating cell nuclear antigen. (PCNA) Assay, Calculate Cell Proliferation Index (PI). The positive rates of P16 protein and P53 protein in 62 lung cancer tissues were 58.1% and 59.7%, respectively. The positive rate of P16 protein in adenocarcinoma was significantly higher than that in small cell carcinoma (P<0.05); the expression of P16 protein in lymph node metastasis positive group was significantly lower than that in negative group (P<0.05); PI grade was grade II P16 protein The expression was significantly higher than that of grade IV (P<0.05). There was no significant difference in the expression of P53 protein in different tissue types of lung cancer. The expression of P53 protein in lymph node metastasis positive group was higher than that in negative group (P<0.01). The expression of P53 protein in different PI grades was significantly higher than that in grade I. (P<0.05) and grade II (P<0.05), grade III was significantly higher than grade I (P<0.05) and grade II (P<0.05). There was no significant correlation between low expression of P16 protein and overexpression of P53 protein. It was revealed that low expression of P16 protein and overexpression of P53 protein all promote the proliferation of lung cancer cells. The expression of P16 protein is related to cell differentiation of lung cancer. Overexpression of P53 protein plays an important role in the metastasis of lung cancer cells. The tumor suppressor gene P53 had no obvious regulation on MTS1/P16 gene. The expression of P53 protein was detected.
