Wig-1是抑瘤基因p53的转录调节靶位之一,编码与基因转录后水平调节相关的锌指结构蛋白。Wig-1几乎在所有类型细胞中均有表达,在脑组织中表达水平最高,在干细胞中表达水平较分化细胞高。Wig-1可结合长双链RNA(dsRNA),也可结合短的dsRNA,如microRNA。Wig-1通过结合p53基因3’非翻译区的富于AU元件稳定p53基因mRNA,正反馈调节p53基因。在此讨论Wig-1与p53功能的关系,在肿瘤、非肿瘤性疾病以及干细胞调控中的作用。
Wig-1 is one of the transcriptional regulatory targets of the tumor suppressor gene p53 and encodes a zinc finger protein involved in the regulation of post-transcriptional gene transcription. Wig-1 is expressed in almost all types of cells, highest in brain tissue, and higher in stem cells than in differentiated cells. Wig-1 binds to long double-stranded RNA (dsRNA) as well as to short dsRNAs such as microRNAs. Wig-1 positively regulates the p53 gene by binding p53-rich mRNA to the AU-rich member of the 3 ’untranslated region of the p53 gene. Here we discuss the role of Wig-1 in relation to p53 function in tumor, non-neoplastic disease and stem cell regulation.
