应用聚合酶链锁反应扩增葡萄糖激酶(GCK)基因微卫星多态标志,GCK-5’,进行GCK基因与国人非胰岛素依赖型(Ⅱ型)糖尿病关联研究,并与非塘尿病组比较.Ⅱ型糖尿病组等位基因4频率明显增高(X~(2)=6.773,P=0.009)。糖尿病组中呈基因型44或4X者明显高于非糖尿病组(x~(2)=6.439,P=0.011)。等位基因频率差异在伴超重/肥胖体重指数亚组比较中亦可见到(x~(2)=7.718,P=0.021)。国人中GCK-5’基因型呈44或4X者,Ⅱ型糖尿病发病风险较基因型呈XX者增加3.6倍左右。结果表明GCK基因与国人Ⅱ型糖尿病关联。
Polymerase chain reaction (PCR) was used to amplify the GCK gene microsatellite marker, GCK-5 ’, to study the association between GCK gene and non-insulin-dependent type 2 diabetes in Chinese population and to compare with non-diabetic group The frequency of allele 4 in type Ⅱ diabetes group was significantly higher than that in type Ⅱ diabetes group (X ~ (2) = 6.773, P = 0.009). The genotype 44 or 4X in the diabetic group was significantly higher than that in the non-diabetic group (x ~ (2) = 6.439, P = 0.011). Allelic differences in allele frequencies were also observed in subgroups with BMI overweight / obesity (x ~ (2) = 7.718, P = 0.021). Chinese people GCK-5 ’genotype was 44 or 4X, type II diabetes incidence of genotype XX was increased by 3.6 times. The results show that GCK gene associated with Chinese type Ⅱ diabetes.
