目的筛选产来氟米特的芽胞杆菌菌株。方法采用液相-四级杆飞行时间质谱的方法分析了31株芽胞杆菌菌株的发酵液胞外成分。结果在谱库扫描得到的代谢物中,来氟米特的含量相对较高,且与谱库的匹配率最高。筛选出含来氟米特成分的菌株为科恩芽胞杆菌(Bacillus cohnii FJAT-10017)、嗜碱芽胞杆菌(Bacillus alcalophilus FJAT-10014)、土壤短芽胞杆菌(Brevibacillus agri FJAT-10018)、解硫胺素解硫胺素芽胞杆菌(Aneurinibacillus aneurinilyticus FJAT-10004)、蜥蜴纤细芽胞杆菌(Gracilibacillus dipsosauri FJAT-14266)。其中该成分含量最高的为蜥蜴纤细芽胞杆菌(Gracilibacillus dipsosauri FJAT-14266),占其发酵液总代谢物相对含量的12.16%。结论蜥蜴纤细芽胞杆菌(Gracilibacillus dipsosauri FJAT-14266)为来氟米特高产菌株。该研究为芽胞杆菌来源的免疫调节剂的开发与利用提供理论依据。
Objective To screen leflunomide-producing Bacillus strains. Methods The extracellular components of the fermentation broth of 31 strains of Bacillus were analyzed by liquid phase-quadrupole time-of-flight mass spectrometry. Results Among the metabolites screened by the library, the content of leflunomide was relatively high and the matching rate with the library was the highest. The selected strains containing leflunomide were Bacillus cohnii FJAT-10017, Bacillus alcalophilus FJAT-10014, Brevibacillus agri FJAT-10018, Aneurinibacillus aneurinilyticus FJAT-10004, and Gracilibacillus dipsosauri FJAT-14266. Gracilibacillus dipsosauri FJAT-14266 was the highest content of this component, accounting for 12.16% of the total metabolites of the fermentation broth. Conclusion Gracilibacillus dipsosauri FJAT-14266 is a leflunomide-producing strain. This study provides a theoretical basis for the development and utilization of Bacillus-derived immunomodulators.
