目的建立中药海藻中总甾醇的含量测定方法。方法采用Bligh-Dyer方法提取总脂,以岩藻甾醇为对照品,磷硫铁试剂为显色剂,利用分光光度法进行含量测定,并进行方法学验证。结果含量测定方法学结果表明,在检测波长576nm下,岩藻甾醇在0.01~0.07g·L~(-1)具有良好的线性关系,r=0.997 0;显色产物在90 min内稳定,RSD 0.54%;平均加样回收率为99.16%,RSD 1.43%(n=6);方法重复性良好,RSD1.05%(n=6)。结论建立的磷硫铁-分光光度法可用于中药海藻中总甾醇的含量测定。
Objective To establish a method for the determination of total sterols in Chinese algae. Methods Bligh-Dyer method was used to extract the total lipids, and fucosterol was used as the reference substance. The phosphorus-sulfur-iron reagent was used as chromogenic reagent, and the content was determined by spectrophotometry. The method was validated by methodology. Results The method of determination of content showed that there was a good linear relationship between the concentration of fucosterol and 0.01 ~ 0.07 g · L -1 at the detection wavelength of 576 nm, r = 0.997 0. The color products were stable within 90 min. The RSD The average recovery was 99.16% and the RSD was 1.43% (n = 6). The reproducibility of the method was good with RSD of 0.05% (n = 6). Conclusion Phosphorus, sulfur and iron spectrophotometry can be used to determine the content of total sterols in Chinese algae.
