[目的]运用微波辅助提取技术提取霍山石斛多糖,并对工艺条件进行优化。[方法]通过单因素实验研究了微波提取霍山石斛多糖过程中物料粒度、提取次数、提取时间、微波功率和料液比对多糖得率的影响;采用正交设计研究了影响多糖得率的关键因素,并与传统回流提取比较。[结果]所有单因素在水平较低时对石斛多糖得率影响较大,水平越高影响越小;正交分析筛选出微波法提取霍山石斛多糖的最优工艺为:提取4次,每次40min,微波功率550 W,并验证了提取次数对多糖得率有显著影响。[结论]微波辅助提取能够提高霍山石斛多糖的提取效率,为开发新的霍山石斛多糖提取技术打下基础。
[Objective] The research aimed to extract polysaccharides from Dendrobium huoshanense by microwave assisted extraction and to optimize the process conditions. [Method] The effects of material size, extraction times, extraction time, microwave power and solid-to-liquid ratio on the yield of polysaccharides were studied by single factor experiments. The key factors affecting yield of polysaccharides were studied by orthogonal design Factor, and compared with the traditional reflux extraction. [Result] All the single factors had a great effect on the yield of D. officinale polysaccharides at lower levels, the smaller the effect was. The optimum extraction process of polysaccharides from D. huoshanense by microwave was as follows: four times, each time 40min, microwave power 550 W, and verify that the number of extraction has a significant impact on polysaccharide yield. [Conclusion] Microwave-assisted extraction could improve the extraction efficiency of polysaccharide from Dendrobium huoshanense and lay a foundation for the development of new extraction technology of polysaccharide from Dendrobium huoshanense.
