目的 探讨术前区域动脉灌注化疗对人结直肠癌细胞增殖的影响。方法 选择术前临床病理证实的结直肠癌患者 20例,于术前选择肿瘤供应动脉行区域灌注化疗, 10 d后行根治手术。免疫组织化学法测定并比较癌组织增殖细胞核抗原( PCNA)及细胞周期蛋白 D1( cyclin D1)的表达水平。同时选择同期未行动脉灌注化疗的结直肠癌患者直接接受根治手术,作为对照组。结果 化疗组及对照组 PCNA标记指数 (LI)分别为 16. 4%± 15. 2%及 38. 1%± 17. 8%( P=0.02) ; cyclin D1 LI分别为 8. 7%± 8. 0% 及 27. 4%± 16. 5%( P=0. 0002)。细胞增殖指标 PCNA及 cyclin D1的表达在化疗组显著低于对照组。结论 术前选择性区域动脉灌注化疗对结直肠癌细胞增殖可能有较明显的抑制作用,从而发挥抑制肿瘤生长的治疗作用。
Objective To investigate the effect of preoperative regional arterial infusion chemotherapy on the proliferation of human colorectal cancer cells. Methods Twenty patients with colorectal cancer confirmed by preoperative clinical pathology were selected. Before the operation, the tumor was supplied with intra-arterial chemotherapy and the radical operation was performed 10 days later. The expression of proliferating cell nuclear antigen (PCNA) and cyclin D1 in cancer tissues was determined and compared by immunohistochemistry. At the same time, colorectal cancer patients who did not undergo arterial infusion chemotherapy during the same period were directly treated with radical surgery as a control group. Results The PCNA labeling index (LI) of chemotherapy group and control group were 16.4%± 15.2% and 38.1%± 17.8% (P=0.02), respectively; cyclin D1 LI was 8.7%± 8 respectively. 0% and 27.4% ± 16.5% (P=0.0002). Cell proliferation markers PCNA and cyclin D1 expression were significantly lower in the chemotherapy group than in the control group. Conclusion Preoperative selective regional arterial infusion chemotherapy may significantly inhibit the proliferation of colorectal cancer cells, and thus play a therapeutic role in inhibiting tumor growth.
