目的:研究新木脂素6-羟基-4-(4-羟基-3-甲氧基苯基)3-羟甲基-5-甲氧基-3,4-二氢(3R,4S)-2-醛基萘(VB-1)诱导人卵巢癌CoC1细胞凋亡作用。方法:体外培养CoC1细胞,碘化丙啶(PI)染色流式细胞术分析细胞凋亡率。比色法测定细胞Caspase-3活性。Western Blot分析Akt蛋白磷酸化水平。结果:VB-1显著增高CoC1细胞凋亡率和激活caspase-3(P<0.05),呈浓度依赖性。1.0μmol/L VB-1以时间依赖方式降低Akt蛋白磷酸化水平(P<0.05)。结论:VB-1能有效诱导CoC1细胞凋亡,其作用机制与抑制Akt蛋白磷酸化相关。
OBJECTIVE: To study the effects of neolignan 6-hydroxy-4- (4-hydroxy-3-methoxyphenyl) 3-hydroxymethyl-5-methoxy-3,4-dihydro (3R, 4S) 2-allenyl naphthalene (VB-1) induces the apoptosis of human ovarian cancer CoC1 cells. Methods: CoC1 cells cultured in vitro, propidium iodide (PI) staining flow cytometry analysis of apoptosis rate. Colorimetric assay of Caspase-3 activity. Western Blot analysis of Akt protein phosphorylation levels. Results: VB-1 significantly increased the apoptosis rate of CoC1 cells and activated caspase-3 (P <0.05) in a dose-dependent manner. 1.0μmol / L VB-1 reduced Akt protein phosphorylation in a time-dependent manner (P <0.05). Conclusion: VB-1 can effectively induce apoptosis of CoC1 cells and its mechanism is related to the inhibition of Akt phosphorylation.
