目的:探讨Ⅰ型神经纤维瘤病(NF1)组织中蛋白激酶B(PKB)、PTEN和c-kit的表达及意义。方法:应用免疫组织化学SP法检测26例NF1、20例孤立性神经纤维瘤和11例正常皮肤组织中磷酸化PKB(p-PKB)、PTEN和c-kit的表达情况。结果:正常皮肤组织中p-PKB,c-kit和PTEN的阳性表达率分别为9.1%,9.1%,100.0%;孤立性神经纤维瘤组织中3个指标的阳性表达率为55.0%,53.9,40.0%;NF1组织中3个指标的阳性表达率为60.0%,65.4%,43.3%。正常皮肤组织中3个指标的表达与后2种组织相比差异均有统计学意义(P<0.05);孤立性神经纤维瘤与NF1组织中3个指标的表达差异无统计学意义(P>0.05)。NF1组织中p-PKB与PTEN的表达呈负相关(r=-0.456,P<0.05);p-PKB与c-kit的表达呈正相关(r=0.462,P<0.05)。结论:p-PKB,PTEN和c-kit可能与NF1的发展关系密切;NF1与孤立性神经纤维瘤的发展机制可能有相同之处。
Objective: To investigate the expression and significance of protein kinase B (PKB), PTEN and c-kit in type Ⅰ neurofibromatosis (NF1). Methods: The expressions of phosphorylated PKB (p-PKB), PTEN and c-kit in 20 cases of solitary neurofibroma in NF1 and 11 cases of normal skin tissues were detected by immunohistochemical SP method. Results: The positive rates of p-PKB, c-kit and PTEN in normal skin tissues were 9.1%, 9.1% and 100.0% respectively. The positive rates of three indexes in solitary neurofibroma were 55.0%, 53.9% 40.0%. The positive expression rates of three indicators in NF1 were 60.0%, 65.4% and 43.3% respectively. The expression of three indexes in normal skin tissue was significantly different from those in the latter two tissues (P <0.05), while there was no significant difference in the expression of three indexes between solitary neurofibroma and NF1 (P> 0.05). There was a negative correlation between the expression of p-PKB and PTEN in NF1 (r = -0.456, P <0.05). There was a positive correlation between the expression of p-PKB and c-kit (r = 0.462, P <0.05). CONCLUSION: The expressions of p-PKB, PTEN and c-kit may be closely related to the development of NF1. The mechanism of NF1 may be similar to that of solitary neurofibroma.
