目的以胶原诱导性关节炎(collagen induced arthritis,CIA)大鼠为研究对象,测定大鼠滑膜血管生成素2(angiopoietin-2,Ang-2)、血管生成素样蛋白2(angiopoietin-like proteins 2,ANGPTL2)表达水平,探讨温化蠲痹方治疗CIA的作用机制。方法选用Wistar大鼠,随机分为造模组(50只)、正常组(10只),造模组于大鼠尾根部注射牛Ⅱ型胶原(typeⅡcollegan of bovine,BCⅡ)乳剂,建立CIA模型。选取30只造模成功大鼠随机分为温化蠲痹方组、甲氨蝶呤组、模型组。给药30 d后,运用实时real time PCR检测大鼠膝关节滑膜Ang-2mRNA、ANGPTL2mRNA的表达。结果温化蠲痹方组和甲氨蝶呤组大鼠滑膜Ang-2 mRNA、ANGPTL2 mRNA表达水平均低于模型组(P<0.05),与甲氨蝶呤组比较差异无统计学意义(P>0.05)。结论温化蠲痹方可有效抑制CIA大鼠滑膜Ang-2mRNA、ANGPTL2mRNA表达,达到抑制血管翳形成而治疗CIA的作用。
Objective To investigate the effects of angiopoietin-like protein 2 (Angiopoietin-2, Ang-2) and collagen-induced arthritis 2, ANGPTL2) expression levels, to explore the role of Wen Wei Hanbi treatment of CIA mechanism. Methods Wistar rats were randomly divided into model group (n = 50) and normal group (n = 10). CIA model was established by injecting modelⅡcollegen of bovine (BC Ⅱ) into the tail of rats. Thirty successful rats were selected and randomly divided into two groups: metformin group, methotrexate group and model group. After administration for 30 days, the real-time real time PCR was used to detect the expression of Ang-2 mRNA and ANGPTL2 mRNA in the synovium of knee joint of rats. Results The expressions of Ang-2 mRNA and ANGPTL2 mRNA in synovial membrane of rats in Wen-hua-bi-fang group and methotrexate group were lower than those in model group (P <0.05), but there was no significant difference compared with methotrexate group P> 0.05). Conclusions Wenhua Jiebi can effectively inhibit the expression of Ang-2mRNA and ANGPTL2mRNA in synovium of CIA rats, and can inhibit the formation of angiogenesis and treat CIA.
